[摘要]目的:研究糖基化终产物(AGEs)对人肾小球系膜细胞半乳糖凝集素3 (galectin3)表达的影响。方法:将培养的人肾小球系膜细胞(HRMC) 用不同浓度(50、100、200及400mg·L-1)的AGEs干预24h或同一浓度(200mg·L-1)的AGEs干预0、24、48及72h,以无血清培养基(DMEM)和相应浓度的牛血清白蛋白(BSA)为对照。用RTPCR检测细胞中半乳糖凝集素3的mRNA水平。结果:不同浓度AGEs组HRMC的半乳糖凝集素3 mRNA水平与对照组相比均有显著差异(P<0.01),且随着AGEs浓度的增高,半乳糖凝集素3 mRNA水平也逐渐增高(P<0.05);AGEs干预HRMC 24、48、72h,半乳糖凝集素3 mRNA水平与对照组相比有显著差异(P<0.05),且随着干预时间的延长,半乳糖凝集素3 mRNA水平也逐渐增高 (P<0.01)。结论:AGEs以时间及剂量依赖的方式促进HRMC半乳糖凝集素3 mRNA的表达。

Effect of advanced glycation end products on the gene expression of galectin3 in cultured human renal mesangial cells

Abstract:Objective To study the effect of advanced glycation end products(AGEs) on the expression of galectin3 in cultured human renal mesangial cells(HRMC). Methods The AGEsBSA was prepared by incubating bovine serum albumin(BSA) with glucose.The cultured HRMC were intervened with the same concentration of BSA and AGEsBSA(50,100,200,400mg· L-1) for 24h or 200 mg· L-1 AGEsBSA for 0,24,48 or 72h respectively. The mRNA expression of galectin3 in HRMC were analyzed by RTPCR.Results Compared with that in cells incubated with DMEM and BSA,the level of galectin3 mRNA in cells incubated with 50,100 or 200,400mg· L-1AGEsBSA respectively for 24h (P<0.01)or with 200mg· L-1AGEsBSA for 0,24,48 or 72h respectively(P<0.05)increased significantly. Moreover, with the increased concentration of AGEsBSA(P<0.05)or the expanded incubating time (P<0.01),the level of galectin3 mRNA increased. Conclusion The AGEsBSA can stimulate the expression of galectin3 in cultured HRMC in a concentration and timedependent manner. ......
