定向诱导兔骨髓间质干细胞分化为神经元样细胞的研究

     摘 要 目的:体外定向诱导兔骨髓间质干细胞(MSC)分化为神经元样细胞。方法:采用FicollPaque液(1.083 g/L)离心分离兔MSC,体外扩增,硫代甘油等试剂的无血清达乐伯克改良必需基本培养基(DMEM)诱导MSC分化为神经元。免疫组化鉴定神经元烯醇化酶(NSE)、神经丝蛋白(NF)、胶质纤维酸性蛋白(GFAP)的表达。结果:兔骨髓间质干细胞在体外扩增原代可获得5×105、10代可获得2×1010个细胞。加入硫代甘油诱导后,MSC胞体收缩,突起伸出;免疫组化显示诱导出的神经元样细胞NSE、NF、表达阳性,GFAP阴性。结论:兔骨髓间质干细胞在体外可以分化为神经元样细胞。

    Induction of the differentiation of rabbits mesenchymal stem cells into neuronlike cells with monothioglycerol

    Abstract Objective:To induce the differentiation of rabbit bone marrow mesenchymal stem cells (rMSC) into neuronlike cells in vitro. Methods: rMSCs were separated from rib marrow with FicollPaque reagent and expanded in culture medium. Differentiation of rMSCs into neurons were induced with DMEM/monothioglycerol. Neuronspecific enolase(NSE),neurofilament (NF),glial fibrillary acidic protein (GFAP) were detected by immunohistochemistry. Results:rMSCs were expanded in culture for 10 passages.After the induction by adding monothioglycerol the MSCs displayed a fibroblastlike morphology. Immunohistochemical assay showed that there were positive expressions of NSE and NFM,on the neuronlike cells,without the expression of the glial astrocyte marker GFAP.Conclusion:It suggests that the differentiation of rMSC into neurons can be induced in vitro. ......
