【Abstract】 AIM: To investigate the effect of diallyl trisulfide coated stents on iNOS protein expression and NO level in the canine models of coronary restenosis, and to explore the mechanisms of diallyl trisulfide coated stents inhibiting neointima formation. METHODS: The diallyl trisulfide coated stent and the control stent were implanted into distal and proximal left circumflex coronary arteries of 20 dogs, respectively. They were sacrificed 1, 3, 10 and 28 d after stenting. The expression of iNOS proteins was assessed by Western blot and the NO level was measured by nitrate reductase. RESULTS: Expression of iNOS proteins was seen on day 1 following the control stent implanting, developed on day 3, peaked on day 10 and lasted till day 28. iNOS expressions at all time points in diallyl trisulfide coated stent group were higher than those in control stent group. Compared with normal blood vessel tissue, the NO production after stenting decreased on day 1, increased on day 3, maximized on day 10, went down to its prestenting level on day 28. At all time points, NO levels in allitridistented sites were higher than those in controlstented sites. CONCLUSION: Diallyl trisulfide coated stent could significantly enhance the iNOS expression and NO production, which may play an important role in inhibiting neointimal hyperplasia. ......
