食管鳞癌P16、P15基因杂合子缺失的研究

     摘 要 目的:探讨P16和P15基因与ESCC发生发展的关系。方法:应用显微切割、PCR、凝胶电泳、AgNO3染色等技术,检测食管鳞癌(ESCC)和高级别不典型增生中P16、P15基因杂合子缺失(LOH),对照正常细胞,分析ESCC和高级别不典型增生组织中LOH的变化,并就LOH率与患者的临床病理参数分别进行单因素分析。结果:ESCC组织中的LOH率为29.7%,高级别不典型增生组织中的LOH率为16.5%。应用SPSS软件对LOH率与患者的性别、组织分化、淋巴结转移进行单因素分析,差异均无显著性(P>0.05)。结论:①从正常鳞状上皮到不典型增生再到癌变的过程中存在基因的异常累积。②P15基因可能与食管鳞癌的发生发展相关。③D9S162位点或其附近可能存在与ESCC发展相关的抑癌基因。

    P16 Protein Expression in Human Hepatocellu

    Abstract Objective:To investigate P16 protein expression ,we infer the role and clinical significance of P16 gene during the occurrence and development of hepatocellular carcinoma. Methods:P16 protein expression was quantitatively determined by im muno-histochemistry in 26 cases of hepatocellular carcinoma.Simultaneously adjacent tumor tissues were contrasted. Results:P16 protein expression in 26 cases of hepatocellular carcinoma was significantly lower than that in adjacent tumor tissues. The expression of P16 protein becomes lower with the increase in grading (P<0.05).Conclusions:Expression absence of P16 gene exist in part of HCC cell,which may be related with developmentand histo - differentiation of HCC.P16 protein may serve as an indicator to predict the prognosis of patients. ......
