GDNF和HSV-GDNF对缺氧时大鼠脊髓运动神经元Bcl-2表达的影响*

     【摘要】 目的 观察缺氧时GDNF和HSV-GDNF对体外培养的大鼠脊髓运动神经元Bcl-2表达的影响。方法 尼氏染色,观察缺氧2~4h和缺氧4h后恢复供氧24、72h脊髓运动神经元存活数。抗Bcl-2血清行免疫组化染色,计数Bcl-2免疫反应(Bcl-2-IR)阳性神经元数目,同时对Bcl-2-IR阳性神经元作平均光密度的色谱分析。结果 GDNF组、HSV-GDNF组脊髓运动神经元缺氧后在神经元存活数、Bcl-2-IR阳性神经元数目和平均光密度三项指标上均明显高于对照组(P<0.05),但HSV-GDNF组的结果更好。结论 HSV-GDNF比GDNF更能增强缺氧时脊髓运动神经元Bcl-2的表达,抑制缺氧后神经元的死亡。

    Effects of GDNF and HSV-GDNF on Bcl-2 expression of spinal cord motoneurons of rats after anoxia in vitro

    【Abstract】 Objective To study the effects of GDNF and HSV-GDNF on Bcl-2 expression of spinal cord motoneurons of rats after anoxia in vitro.Methods Spinal cord motoneurons were Nissl stained and counted the number of survival neurons after anoxia 2,4 hours and reoxygenation for 24 and 72 hours.At the same time,neurons were immunohistochemical stained by the antiserum of Bcl-2.The number and the mean optical densities of Bcl-2 immunoreactive positive neurons were respectively observed.Results By drawing a comparison with corresponding samples,it showed that these three items in GDNF and HSV-GDNF group were significantly improved after anoxia.The effects of HSV-GDNF were better than those of GDNF after anoxia.Conclusion HSV-GDNF may protect motoneurons from the damage induced by anoxia in better degree than GDNF and improve Bcl-2 expression of spinal cord motoneurons. ......
