奥曲肽对胃癌细胞株SGC7901生长调控作用的研究

     [摘要]目的研究奥曲肽(OCT)对人胃中分化腺癌细胞株SGC7901体外生长抑制作用的机制。方法MTT法测定OCT对SGC7901细胞生长的调控;流式细胞术分析OCT对SGC7901细胞周期的影响;免疫细胞化学染色法检测表皮生长因子受体(EGFR)的表达;放射免疫法测定细胞培养上清液中表皮生长因子(EGF)和胃泌素的含量。结果OCT对SGC7901细胞生长有抑制作用;OCT可诱导SGC7901细胞G0/G1期阻滞,加药后24小时最显著,同时伴S期细胞比例减少; OCT作用后24、36小时,SGC7901细胞EGFR表达较对照组明显减少。加入OCT24小时,细胞培养上清液中EGF和胃泌素含量明显下降。结论OCT可通过多种途径抑制体外培养的SGC7901细胞的增殖。

    Experimental study of regulatory effect of octreotide on human gastric cancer cell line SGC7901 growth in vitro

    [Abstract]ObjectiveTo study the mechanisms that regulatory effect of somatostatin analogue(octreotide,OCT)on the growth of gastric cancer cell line SGC7901 in vitro.MethodsCell proliferation was evaluated by means of MTT assay; cell cycle distribution was measured by flow cytometric analysis . The immunocytochemical stain and image analysis were applied to measure the expression of EGFR protein.The contents of EGF and gastrin were determined by radioimmunoassay (RIA). ResultsThe growth of human gastric cancer cell line SGC7901 was inhibited by OCT.OCT also induced SGC7901 arrest at G0/G1 phase and S phase decrease, which peaked at 24h. OCT regulated downwards the expression of EGFR. At 24h,the contents of EGF and gastrin were significantly decreased by OCT. ConclusionsOCT inhibits the growth of gastric cancer cell line SGC7901 by many pathways in vitro. ......
