固相萃取高效液相色谱质谱法测定血浆中乙醇醛、甘油醛的含量

     摘 要 采用柱前衍生、固相萃取、高效液相色谱质谱联用方法,建立了血浆中乙醇醛(GCO)、甘油醛(GCE)含量的测定方法。衍生试剂为3methyl2benzothiazolinonehydrazone hydrochloride(MBTH);内标物为3羟基丁醛;HPLC流动相为甲醇醋酸铵(5 mmol/L),梯度洗脱;质谱电喷雾选择离子检测。比较了糖尿病大鼠和正常大鼠血浆中GCO、GCE的含量,正常大鼠血浆中GCO和GCE的含量分别为0.0574±00036 μmol/L、00186±0.0019 μmol/L; 糖尿病大鼠血浆中GCO和GCE的含量分别为0.0763±0.0071 μmol/L、 0.0240±0.0023 μmol/L,差异显著。

    Abstract Diabetes is a serious disease that endangers people health. Many factors are considered to be related to diabetes and its complications; one of those is that the saccharides react with amino compounds to form advanced glycation and oxidation end products (AGEs). In this study, glycolaldehyde (GCO) and glyceraldehydes (GCE) in rat plasma were studied. Firstly, they were transformed into their derivatives that can be detected conveniently, and then with solid phase extraction and concentration, the derivatives were determined with high performance liquid chromatographymass spectrometry. Mobile phase was MeOH and CH3COONH4 (5 mmol/L), gradient condition was used. Proper conditions of mass spectrometry was chosen so that only 3methyl2benzothiazolinonehydrazonehydrochloride (MBTH)GCO, MBTHGCE and MBTHC4H8O2 were detected. The GCO and GCE in normal rat plasma is (0.0574±0.0036) μmol/L and (0.0186±00019) μmol/L respectively; the GCO and GCE in diabetic rat plasma is (0.0763±0.0071) μmol/L and (0.0240±0.0023) μmol/L respectively.
