微电极阵技术对多非利特干预豚鼠心室肌电生理特性的应用研究

     摘要: 目的: 探讨微电极阵(Microeletrode arrays, MEA)记录技术记录心肌组织及细胞群动作电位和电激动传导的作用。研究新型Ⅲ类抗心律失常药多非利特不同浓度梯度对豚鼠心室肌场电位时程(field action potential duration,fAPD )及心率的影响。方法: 随机将40只豚鼠分为对照组、胺碘酮组、10-7mol/L多非利特组、及10-6mol/L多非利特组,应用MEA记录技术,从60个位点标测心室肌场电位(field potentials, FPs)波形,监测多非利特对心室肌fAPD及心率的影响,并与经典的Ⅲ类抗心律失常药胺碘酮进行对比研究。结果:MEA记录技术可以记录到60个位点心室肌FPs不同的波形;多非利特可明显延长豚鼠心室肌fAPD及减慢心率,不同浓度多非利特间差异有统计学意义(P<0.05),但多非利特组与胺碘酮组对比对豚鼠心室肌fAPD及心率影响差异无统计学意义(P>0.05)。结论:MEA是记录心肌组织及细胞群动作电位和电激动传导的新技术 ,同时是抗心律失常药物组织水平疗效监测的理想工具。

    Study on the effect of dofetilide on ventricular myocardial electrophysiological property by MEA

    Abstract:Objective: The morphology of field potentials (FPs) were studied by microelectrode arrays (MEA) mapping technique. The effects of amiodarone and dofetilide on ventricular myocardial field action potential duration (fAPD) and heart rate were mapped by MEA in guinea pigs. Methods: The morphology of field potentials (FPs) was mapped by a microelectrode arrays (MEA) recording techniques in eighty guineapigs. The eighty guineapigs were randomly divided into four groups: control group, amiodarone group, 10-6 mol/l and 10-7 mol/l dofetilide group. Both ventricular muscle field potential (FPs) and fAPD were observed by MEA. Results: These results showed that MEA could record sixty different shapes of fAPDs and ventricular activation sequence. The fAPD of ventricular myocytes was remarkably prolonged and the heart rate was reduced in both amiodarone and dofetilide group (P<0.05), still clear difference of fAPD was produced between 10-6 mol/l and 10-7 mol/l dofetilide groups by (P<0.05). Conclusions: Thus MEA is an idea tool in investigating new antiarrhythmia drugs and proarrhythmia mechanism of antiarrhythmia drugs. ......
