离子液体双水相体系萃取分离牛血清白蛋白研究

     摘 要 建立了由亲水性离子液体四氟硼酸1甲基3丁基咪唑([Bmim]BF4 )和KH2PO4 形成的双水相体系萃取分离牛血清白蛋白(BSA)的新方法。研究了不同盐及盐的浓度、离子液体浓度以及蛋白质用量、溶液酸度、其它共存物质对双水相成相及BSA萃取率的影响,结果表明, 磷酸二氢钾盐浓度为80 g/L,离子液体浓度在160~240 mL/L,BSA的浓度为30~50 mg/L,溶液酸度在pH 4~8范围,离子液体双水相体系对BSA有较高的萃取率。用加入不同类型表面活性剂探讨了离子液体与蛋白质之间的作用。

    Ionic Liquid Aqueous Twophase System

    Abstract A new method to extract and separate Bovine Serum Albumin(BSA) in a new aqueous twophase system(ATPS) formed by a room temperature watersoluble ionic liquid [Bmim]BF4 (1butyl3methylimidazoliumtetrafluoroborate) and phaseforming salt KH2PO4 was studied. The effects of concentration of ionic liquid, salt and BSA, solution pH value and other coexistence sulstance on the extraction efficiency and the formation twophase were investigated systematically. With the solution pH value 4-8, the addition of BSA 30-50 mg/L, [Bmim]BF4 160-240 mL/L and KH2PO4 80 g/L, Ionic liquidATPS had the highest extraction efficiency.With different surfactant, the interaction of protein and ionic liquid were discussed initiatively. ......
