     Effects of 17β estradiol on apoptosis and mRNA expression of MT1MMP in endothelial cell under high glucose condition

    【Abstract】 AIM: To explore the effect of 17β estradiol (E2)on apoptosis and membranetype matrix metalloproteinase 1(MT1MMP) mRNA expression in endothelial cell line EVC304 under the high glucose condition 30 mmol/L. METHODS: The proliferation of EVC304, which is treated by high glucose(30 mmol/L) and/or E2 of different concentrations respectively, was measured by MTT assays. Then the apoptosis of ECV304 was detected by flow cytometry(FCM) and DNA ladder assays. Finally, MT1MMP mRNA expression was measured by semiquantitative RTPCR. RESULTS: After ECV304 had been treated by high glucose(30 mmol/L) ,its proliferation and mRNA expression of MT1MMP was inhibited significantly(P<0.01). However, the tendency fall back after E2 had been administrated(P<0.01). Furthermore, this effect of E2 reached a peak at its concentration from 0.01 to 0.10 mmol/L. The apoptosis was only detected in cells which had been administrated by high glucose and E2 at low (0.001 nmol/L) and high (1.0 nmol/L) concentration by DNA Ladder assays. CONCLUSION: High glucose has a strong toxicity on vascular endothelial cells. Moderate level of E2 has a protective effect on vascular endothelial cells. E2 can enhance MT1MMP expression of endothelial cells under high glucose condition. ......
