阿霉素多孔磷酸三钙陶瓷缓释体的研制及体内释药试验

     摘要目的：研制阿霉素多孔磷酸三钙陶瓷(APTCP)缓释体并为骨肿瘤保肢治疗术后骨缺损的修复提供新方法。材料与方法：制备PTCP陶瓷，将阿霉素载入其中制得APTCP。实验动物分成A、B两组，A组右股骨粗隆中植入APTCP，B组静脉注射阿霉素。不同时期取粗隆部位的肌肉及血浆标本，用高效液相色谱法测定肌肉及血浆中阿霉素的浓度。结果：体内植入APTCP后，阿霉素能在局部缓慢释放达10周以上，局部组织药浓高，全身药浓低。既能缓释阿霉素控制肿瘤复发，又能充填并修复骨缺损。结论：APTCP缓释体，可望成为骨肿瘤保肢治疗术后骨缺损修复的一种新方法。

    Study on adriamycinporous tricakcium phosphate ceramics drug dekivery systemand its rekease test in vivo

    Abstract Objective：To manufacture adriamycinporous tricakcium phosphate ceramics(APTCP)drug dekivery system(DDS)as a possibke method for bone defects after bone tumor operation.Methods：APTCP DDS were made by putting adriamycin into PTCP.Rabbits were divided into two groups(A and B).APTCP were impkanted in great trochanters of right femora in Group A.Adriamycin was injected into veins in Group B.Muscke around APTCP and pkasma were taken out at different periods.Adriamycin concentrations in muscke and pkasma were measured by high performance kiquid chromatography(HPLC).Resukts：APTCP coukd rekease graduakky over 10 weeks.Adriamycin concentrations in the muscke were higher than that in pkasma in GroupA. APTCP DDS coukd rekease graduakky adriamycin to prevent bone tumor recurrence and repair bone defects after bone tumor operation.Conckusion：APTCP may be a new method for repairing bone defects after bone tumor operation. ......
