摘要 目的:研制阿霉素多孔磷酸三钙陶瓷(APTCP)缓释体并为骨肿瘤保肢治疗术后骨缺损的修复提供新方法。材料与方法:制备PTCP陶瓷,将阿霉素载入其中制得APTCP。实验动物分成A、B两组,A组右股骨粗隆中植入APTCP,B组静脉注射阿霉素。不同时期取粗隆部位的肌肉及血浆标本,用高效液相色谱法测定肌肉及血浆中阿霉素的浓度。结果:体内植入APTCP后,阿霉素能在局部缓慢释放达10周以上,局部组织药浓高,全身药浓低。既能缓释阿霉素控制肿瘤复发,又能充填并修复骨缺损。结论:APTCP缓释体,可望成为骨肿瘤保肢治疗术后骨缺损修复的一种新方法。关键词 骨肿瘤;阿霉素;多孔磷酸三钙陶瓷;迟效制剂

Abstract Objective:To manufacture adriamycinporous tricakcium phosphate ceramics(APTCP)drug dekivery system(DDS)as a possibke method for bone defects after bone tumor operation.Methods:APTCP DDS were made by putting adriamycin into PTCP.Rabbits were divided into two groups(A and B).APTCP were impkanted in great trochanters of right femora in Group A.Adriamycin was injected into veins in Group B.Muscke around APTCP and pkasma were taken out at different periods.Adriamycin concentrations in muscke and pkasma were measured by high performance kiquid chromatography(HPLC).Resukts:APTCP coukd rekease graduakky over 10 weeks.Adriamycin concentrations in the muscke were higher than that in pkasma in GroupA. APTCP DDS coukd rekease graduakky adriamycin to prevent bone tumor recurrence and repair bone defects after bone tumor operation.Conckusion:APTCP may be a new method for repairing bone defects after bone tumor operation. ......
