5-氟尿嘧啶脂质体的制备与评价

     【摘要】 目的 改进5-氟尿嘧啶(5-Fu)脂质体的处方和制备工艺,提高脂质体的包封率及稳定性。方法 采用逆相蒸发法制备5-氟尿嘧啶脂质体,优化脂质体处方;考察脂质体的形态、粒径、粒径分布、包封率、Zeta 电位、体外释药等特性以及脂质体的稳定性,对脂质体进行质量评价。结果 所制得的脂质体外观较好,粒径均匀;空白脂质体平均粒径460.5nm,多分散性指数0.862,Zeta电位-4.43mV;5-氟尿嘧啶脂质体平均粒径221.3nm,多分散性指数0.644,Zeta电位-7.44mV;5-氟尿嘧啶脂质体包封率达到40.5%,体外释药起到缓慢释药的作用,且稳定性良好。结论 脂质体达到设计要求。

    【Abstract】 Objective To optimize the formulation and technology of 5-Fu liposomes,evaluate and improve the encapsulation efficiency and stability of 5-Fu liposomes.Methods The liposomes were prepared by reverse-phase evaporation technique.On the base of the single factor investigation,preparation conditions of 5-Fu liposomes were optimized.The Zeta potentials and the polydispersity were evaluated,the stabilities and the release characteristics in vitro were investigated.Results The liposomes were spherical vesicles with uniform sizes.The mean diameter of the blank liposomes and the 5-Fu liposomes were 460.5nm and 220.1nm ......
