虎杖苷对烧伤血清和LPS作用下大鼠平滑肌细胞蛋白激酶C活性的影响

    【Abstract】 AIM: To explore the mechanism of polydatin (PD) treating shock through studying the influence of PD on the activity of protein kinase C (PKC) of vascular smooth muscle cells (VSMC) when treated by sera from burned rats and lipopolysaccharide (LPS). METHODS: After VSMC was treated by sera from burned rats or LPS, the activity of PKC was measured by γscintillation counting instrument. RESULTS: Activity of PKC of VSMC cytoplasm after treated by sera from burned rats decreased (P<0.05 vs normal group), but activity of PKC of cell membrane increased (P<0.05 vs normal group). When VSMC was treated with PD, the activity of PKC of VSMC cytoplasm increased and that of cell membrane decreased (P<0.05 vs burned group and control group, P>0.05 vs normal group). However, the activity of PKC of VSMC cytoplasm and cell membrane treated by LPS increased (P<0.05 vs normal group), after PD treated VSMC the activity of PKC decreased again (P<0.05 vs LPS group and control group, P>0.05 vs normal group). CONCLUSION: PD antagonizes the damage effects of LPS and sera of burned rats by reversing the PKC activities, which may be one of the molecular mechanisms of PD antishock property. ......
