【Abstract】 AIM: To study the method of tracking of allogenically grafted rat bone marrow stem cells (BMSCs) labeled with Feridex in rat liver. METHODS:Feridex was used to label isolated BMSCs. Labeled BMSCs were examined by Prussian blue staining and electron microscopy. Successfully labeled BMSCs (2.0 mL,1×109/L) were respectively transplanted through portal vein, local liver parenchyma and caudal vein of the SD rats. Six hours before transplantation and 6 h, 1, 3, and 5 weeks after transplantation, magnetic resonance imaging (MRI) examination (the sequences used were SET2WI, FSET2WI, GRET2WI) was conducted on rat livers. RESULTS: Prussian blue staining confirmed the labeling efficiency was above 90%, and transmission electron microscopy showed numerous iron particles in the cytoplasm of Feridexlabeled BMSCs. MRI (SET2WI) showed: no remarkable low signal change was found in the liver images before transplantation of all 3 groups. In the porta hepatis of portal vein transplantation group remarkable low signal changes were seen 6 h after transplantation and the extent gradually expanded 1 and 3 weeks after transplantation, while the low signal change could not be seen 5 weeks after transplantation. In the section where the cells were injected in of liver topical transplantation group remarkable low signal changes were seen 6 h after transplantation and the extent gradually expanded 1, 3 and 5 weeks after transplantation. No remarkable low signal change was found in the liver images of caudal vein transplantation group at all time points after transplantation. CONCLUSION: Allogenically grafted Feridexlabeled BMSCs can be tracked effectively in rat liver by MRI. ......
