参麦注射液对心肌缺血再灌大鼠内皮细胞功能及结构的影响

     摘要:目的 研究参麦注射液(SM)对心肌缺血再灌注损伤(IR)大鼠内皮细胞的保护作用。方法 实验大鼠36只,随机分为3组:假手术对照组(Ⅰ);心肌缺血再灌组(Ⅱ);心肌缺血再灌+SM组(Ⅲ)。观察各组缺血30min再灌注3h后血循环内皮细胞数(CEC)、一氧化氮(NO)、内皮素(ET)的含量变化,同时电镜观察冠脉内皮细胞的超微结构。结果 ①与假手术组比缺血再灌组CEC增加(P<0.01),NO下降(P<0.05),ET含量增加(P<0.01),内皮细胞的超微结构受损。②SM干预后CEC下降、NO上升明显(P<0.05),ET显著减少(P<0.01),内皮细胞的超微结构受到保护。结论 IR可诱发内皮功能紊乱,而SM能改善MIR损伤时的内皮形态及功能,对心肌具有保护作用。

    Effects of Shenmai on Endothelial Cell in Myocardial IschemiaReperfusion in Rats

    ABSTRACT:Objective To study the protective effects of Shenmai (SM) on endothelial cell in myocardial ischemic reperfusion (MIR) in rats.Methods 36 rats were randomly divided into 3 groups(n=12 in each):sham operated group(Ⅰ), MIR group(Ⅱ) and SM group(Ⅲ). The number of circulation endothelial cells (CEC), plasma endothelin (ET),nitric oxide (NO) were measured after 30 minutes ischemia and 3 hours reperfusion , and the ultrastructure changes in arterial endothelial cells were observed under electron microscope. Results ①Comparing with group Ⅰ,the number of CEC increased (P<0.01),NO decreased(P<0.05)and ET increased (P<0.01)in groupⅡ and there were abnormal changes of the endothelial ultrastructure . ②Shenmai reversed the results of the above mentioned indices markedly.Conclusion Reperfusion results in dysfunction of endothelium (NO level decreasing and ET level increasing ), Shenmai reduces myocadial ischemioreperfusion injury by protecting endothelial structure and function. ......
