丝裂霉素对人胃癌细胞谷胱甘肽转移酶表达的影响

     【摘要】 目的:探讨丝裂霉素对胃癌细胞谷胱甘肽转移酶(Glutathione Strans ferase,GSTπ)表达的影响。方法:利用人胃癌细胞株BGC823进行培养,加入丝裂霉素后5h和10h后采集细胞,进行免疫组化染色。结果:5h组,胞质中GSTπ含量下降,胞核中GSTπ增多;10h组其胞质与胞核中仍有少量GSTπ表达。结论:丝裂霉素对BGC823癌细胞有明显的杀伤作用,其途径之一可能与抑制GSTπ基因表达有关。

    Influence of Mitomycin on Expression of GSTπ in Human Gastric Cancer Cells

    【ABSTRACT】 Objective:To study the killing function of mitomycin on gastric cancer cells and the expression of GSTπ . Methods: As human gastric cancer cells were cultured, mitomycin were injected to culture continuously for 5h or 10h. The gastric cancer cells were observed under optical microscope and transmission electron microscope after staining with immunohistochemical method.Results: The morphology of gastric cancer cell was not changed significantly, but content of GSTπ decreased in the 5 hours group in the group. The changes in the 10 hours group were as follows: the cell organ were obviously destroyed, the cell membrane were burst, the expression of GSTπ was little in cytoplasm and nuclear. Conclusion:Mitomyclin has a killing fuction to gastric cancer cells, one of manners was related to restraining the expression of GSTπ gene. ......
