siRNA对胃腺癌细胞株SGC-7901血管内皮生长 因子基因表达的作用

     [摘要]目的 研究siRNA对人胃腺癌细胞株SGC-7901血管内皮生长因子(VEGF)基因表达的影响。方法利用体外合成带有T7启动子的VEGF基因DNA片段,转录针对VEGF mRNA的siRNA,通过脂质体2000将合成的siRNA转染入SGC-7901细胞,设置转染VEGF错义序列组、脂质体组、空白对照组作为对照,用MTT法检测细胞抑制率,流式细胞仪检测细胞周期的改变,RT-PCR检测转染后VEGF mRNA表达水平的变化,ELISA法检测细胞培养液中VEGF蛋白分泌量的变化。结果 所设计的两个靶位点siRNA均能有效抑制胃腺癌细胞的生长,并使细胞周期阻滞于G0 /G1 期;VEGF mRNA的表达也受到有效抑制,明显减少;同时,对应的VEGF蛋白水平也显著降低,作为阴性对照的错义序列组siRNA则没有出现这种变化。结论 体外转录合成的siRNA可抑制胃腺癌SGC-7901细胞VEGF基因的表达。

    [ABSTRACT]ObjectiveTo evaluate the gene expression of VEGF by RNA interference in human gastric cancer. Methods siRNAs targeting VEGF mRNA was obtained by in vitro transcription, which was transfected into human gastric cancer cell lines SGC-7901 with Lipofectamine 2000. The non-transfected cells, cells treated with Lipofectimine and cells treated with SCR were taken as controls. MTT was used to detect the inhibitive rate of cell growth.
