静脉注射羟基喜树碱脂肪乳的制备

    摘要 目的 制备静脉注射羟基喜树碱脂肪乳。方法 采用高压均质法制备羟基喜树碱脂肪乳。以包封率、离心稳定性参数、粒径、药物含量等为评价指标,通过正交设计对处方进行优化,在最佳处方的基础上,考察了工艺参数对乳剂的影响,确定了较优的工艺过程。结果 在优化的处方和工艺条件下,所制备的羟基喜树碱脂肪乳平均粒径为176 nm,包封率为97.6%。结论 该处方和工艺制得的羟基喜树碱脂肪乳性质稳定、重复性好,可满足注射的要求。

    Preparation of hydroxycamptothecin lipid emulsion for intravenous administration

    Abstract Objective To prepare hydroxycamptothecin (HCPT) lipid emulsion for intravenous administration. Methods The HCPT lipid emulsion was obtained by a high-pressure homogenization. The physical and chemical stability of emulsion were evaluated with incorporation ratio, the stability parameters of centrifugation, content of HPCT, appearance of emulsion and droplet size. The optimum formulation was established by orthogonal experimental design, and processing factors were optimized based on the optimized formulation. Results The volumn-weighed mean droplet size of HPCT lipid emulsion was 176nm, the entrapment rate was 97.6%. Conclusions The HPCT lipid emulsion with good and stable quality made by this optimum formulation and preparation technique was fitted to be used for intravenous administration.
