摘要:目的探讨灰树花子实体多糖中 β葡聚糖含量的测定方法。方法多糖粗提物依次用高温 α淀粉酶、木瓜蛋白酶和 α1,4葡萄糖水解酶水解,通过测定其中葡萄糖的含量计算β葡聚糖的含量。结果确定了测定方法中几种酶的用量。在0~300 mg·L-1范围内线性关系良好,测得灰树花子实体多糖中 β葡聚糖含量为3.26%~3.51%,3组样品的平均回收率为90.5%。结论该方法操作简便,准确度高。关键词:灰树花; 多糖 β-葡聚糖; 酶
Enzymatic Measurement of βglucans in Polysaccharides of Grifola frondosa

Abstract:ObjectiveAn enzymatic method for analysis of βglucan in polysaccharides of Grifola frondosa has been studied. MethodsAfter hydrolyzing by thermostable αamylose, papain and amyloglucosidase, the value of glucose were measured and the content of βglucan were computed .ResultsThe dosage of several enzymes were determined and the content of βglucan in the samples were in the range of 3.26%~3.51%. The average recovery was 90.5%. ConclusionThe enzymatic method is simple and with high accuracy. ......
