     摘要：目的采用高效液相色谱蒸发光散射(HPLCELSD)法建立柴胡中柴胡皂苷a及柴胡皂苷d的含量测定方法，并测定不同产地柴胡中柴胡皂苷a，d的含量。方法采用 Phnomenex C18 柱(250 mm×4.6 mm，5 μm ), 乙腈水(43∶57)为流动相, 流速0.8 ml/min，Alltech ELSD2000ES蒸发光散射检测器,漂移管温度99℃，气速3.0 L/min, 外标法定量。结果柴胡皂苷a 在0.296～2.96 μg范围内呈良好线性关系, 回归方程为Ya=1.387 6X-2.920 2(r=0.999 8),平均回收率为97.4%,RSD为2.39%(n=6)；柴胡皂苷d 在0.299～2.99 μg范围内呈良好线性关系,回归方程Yd=1.259 5X-1.290 6(r=0.999 6),平均回收率为98.1%,RSD为2.47% (n=6)。结论该方法简便易行,结果准确,重现性好，可作为柴胡药材质量控制的方法。

    Determination of Saikosaponin a and d in Radix Bupleuri by HPLCELSD

    Abstract：ObjectiveTo establish a determination method of saikosaponin a and d in Radix Bupleuri using HPLCELSD.MethodsPhnomenex C18 column (250 mm×4.6 mm ,5μm) was used.The mobile phase was a mixture of acetonitrile-water(43∶57).The flow rate was 0.8 ml/min. Alltech ELSD2000ES evaporative light scattering detector was used as detector. Drift tube temperature was 99℃.The gas flow was 3.0 L/min.The external standard method was used.ResultsSaikosaponin a was linear in the range of 0.296～2.96 μg. The regression equations was Ya=1.3876X-2.9202(r=0.9998).The average recovery was 97.4%( RSD = 2.39%, n=6);Saikosaponin d was linear in the range of 0.299～2.99μg. The regression equations was Yd=1.259 5X-1.290 6(r=0.999 6).The average recovery was 98.1% ( RSD = 2.47% ,n=6). Conclusion This method was simple and easy to perform and the results obtained were accurate and reproducible.It can be used for the quality control of Radix Bupleuri. ......
