四物汤及其三个加味方对大鼠急性血淤模型的药效作用比较

     摘要:目的观察比较四物汤及其加味方对大鼠血淤模型的药效作用。方法选用Wistar大鼠48只,随机分为正常对照组、血淤模型组、四物汤组、圣愈汤组、桃红四物汤组、胶艾汤组,每组8只。各组大鼠连续灌胃7 d,第8天除正常组外,其余5组大鼠同时皮下注射盐酸肾上腺素,按0.9 mg/kg剂量,2次/d,2次给药之间进行1次冰水浸泡刺激,时间为5 min,处置后停食。18 h后用乙醚麻醉剖腹,腹主动脉采血,作血液流变学检测及凝血时间的测定。结果与血淤模型组比较,四物汤组和圣愈汤组的大鼠各切变率下的血液黏度均明显降低(P<0.05或P<0.01或P≤0.001);桃红四物汤组在切变率1 s-1下的血液黏度降低有显著性意义(P<0.05)。与血淤模型组比较,圣愈汤组和桃红四物汤组的血浆黏度明显降低(P<0.05)。与血淤模型组比较,桃红四物汤组和胶艾汤组大鼠红细胞聚集率均有明显降低(P<0.001)。与四物汤组比较,圣愈汤组、桃红四物汤组和胶艾汤组大鼠红细胞聚集指数均有明显降低(P<0.05或P<0.01),其中桃红四物汤组和胶艾汤组大鼠的红细胞聚集指数降低非常明显(P<0.01)。与血淤模型组比较,四物汤组、圣愈汤组和桃红四物汤组大鼠红细胞压积均明显降低(P<0.05或P<0.01)。与血淤模型组比较,四物汤组及其三个加味方的凝血时间均明显延长(P<0.01或P<0.01)。结论 四物汤及其三个加味方作用于血淤模型均能显示出一定活血作用,活血作用的强度因配伍而有所不同,四物汤配伍活血、益气药时的活血作用较强。提示方剂的效用与其所作用的对象的状态有关。

    Function Comparison between Siwu Tang and its Three Additional Prescriptions` Medicine on Rats Acute Blood Stasis

    Abstract:ObjectiveObserve and compare Siwu Tang and its additional prescriptions' medicine function on the rat blood stasis model.MethodsChoose 48 Wistar rats and divide into normal group, blood stasis model group, Siwu Tang group, Shengyu Tang group, Taohong Siwu Tang group, Jiaoai Tang group at random, 8 rats in per group. Each rat was continuously clystered for 7 days. On the 8th day except normal group, rest 5 rats groups received the subcutaneous injection of Adrenaline Hydrochloride, dose was 0.9 mgs/kg, 2 times/d. The rats were ice water soaked as stimulation for 5 mins between 2 injections. Stop food after stimulation. After 18 h, cut the stomach by etherization. Get the blood from abdominal aorta to make examination of hemorheology and the measurement of the clotting time.ResultsCompared with the blood stasis model group, in the rats of group Siwu Tang and Shengyu Tang, the blood viscosity lowered obviously under the shear rate(P<0.05 or P<0.01 or P≤0.001); In Taohong Siwu group under shear rate 1 s-1, blood viscosity lower had very obvious meaning (P<0.05). Compared with the blood stasis model group, the rat's Erythrocyte aggregations rate of Taohong Siwu Tang and Jaoai group lowered obviously(P<0.001). Compared with the group of Siwu Tang, Shengyu Tang group, Taohong Siwu group and Jaoai group's rats' erythrocyte aggreagation index numbers all lowered obviously(P<0.05 or P<0.01). Among them, in the Taohong Siwu Tang and Jiaoai, the index number rats' Erythrocyte aggregation lowered very obviously(P<0.01). Compared with the blood stasis model group, in the set of Siwu Tang, the set of Shengyu Tang Taohong Siwu, rats' H'crit all lowered obviously(P<0.05 or P<0.01). Compare with the blood stasis model set, the set of Siwu Tang and its additional prescriptions' clotting time all prolonged obviously(P<0.01 or P<0.01).ConclusionSiwu Tang and its additional prescriptions can all show certain function of promoting blood flow on blood stasis model. The power of the promoting blood flow function has the dissimilarity because of the compatibility. When Siwu Tang and its compatibility work on the promoting blood flow, benefiting vital energy, it has stronger function on promoting blood flows reminding, the effect of prescription is relevant with the status of the object. ......
