肾阳虚肺纤维化和肺纤维化大鼠肺组织糖皮质激素受体的表达

     摘要:目的探讨糖皮质激素受体(GR) mRNA在肾阳虚肺纤维化和肺纤维化大鼠肺组织的表达及其意义。方法60只健康SD大鼠,随机分为生理盐水组(NS)、肺纤维化组(BLM)及肾阳虚肺纤维化组(RYA),每组再分成两亚组,每亚组10只,用博莱霉素气管内滴入及腺嘌呤灌胃复制肺纤维化及肾阳虚肺纤维化大鼠模型,于14 d及28 d取大鼠肺组织,运用组织芯片,通过HE及胶原纤维染色以判断肺炎症及纤维化程度和对肺内胶原蛋白定量,通过原位杂交(ISH)对肺组织GRmRNA表达进行定位定量分析。结果14 d时,肾阳虚组及肺纤维化组GRmRNA表达皆呈上调趋势,但无统计学意义(P>0.05);28 d时,BLM组GRmRNA表达明显上调,与BLM组比较,RYA组GRmRNA表达则明显下调(P<0.05)。结论肺组织GRmRNA表达的上调是糖皮质激素防治肺纤维化之所以有效的机理之一,而肾阳虚状态下肺纤维化肺组织GRmRNA表达下调是临床上许多肺纤维化病人对糖皮质激素不敏感的原因之一,同时,温阳药也极可能具有防治肺纤维化的作用。

    GR mRNA Expression in Lung Tissues in Pulmonary Fibrosis with Renalyang Asthenia and Pulmonary Fibrosis Rat Models

    Abstract:ObjectiveTo investigate the glucocorticoid receptor(GR)mRNA expression in lung tissues in pulmonary fibrosis with renalyang asthenia and pulmonary fibrosis rat models and its clinical value.Methods60 healthy SpragueDawley rats were randomly divided into the NS group(NS),the pulmonary-fibrosis group(BLM) and the pulmonary fibrosis with renal-yang asthenia group(RYA),every group had 20 rats and were subdivided into two subgroups by 14 days and 28 days courses,each subgroup had 10 rats.Pulmonary fibrosis rat models were established by blemycin-induced by way of oncedripping into trachea, in addition, pulmonary fibrosis with renalyang asthenia rat models as well as adding adenine by primed-stomach once each day,on day 14 and day 28 respectively,these rats were killed and the rat lung tissues were acquired;and by means of tissue microarray associated with computer-picture analysis technology,the severe degree of inflammation and fibrosis,collagen quantity in rat lungs,were estimated by HE and collagen dye, GR mRNA expression were detected by in situ hybridization way.ResultsGR mRNA expression in rat lung of BLM were upregulated(P﹤0.05),however,those in RYA were downregulated(versus BLM P﹤0.05) on day28; GR mRNA expression among three groups were not significant on day14.ConclusionGR mRNA upregulation in lung tissue could be one of the mechanism of glucocorticoid's effective cure for pulmonary fibrosis,and GR mRNA downregulation in lung tissue in renalyang asthenia syndrome could be one of cause of glucocorticoid's noneffective therapy for pulmonary fibrosis,in addition,the Chinese traditional medicine with warming Yang effect could be effective therapy for pulmonary fibrosis. ......
