微囊藻毒素LR促肝癌过程p53、p16基因mRNA表达

     摘要:目的 探讨微囊藻毒素LR(MCLR)促肝癌的分子机制。方法 采用二阶段致癌理论建立中期试验动物模型,γ谷氨酰转肽酶(γGT)染色检验MCLR的促癌作用,并以RTPCR结合图像分析技术精确测定大鼠肝脏p53和p16基因的表达情况。 结果 (1)MCLR在促大鼠肝癌过程中能显著增加γGT阳性率。(2)MCLR在促大鼠肝癌过程中能显著增加大鼠肝脏p53 mRNA表达强度,而对p16 mRNA表达强度没有影响。结论 调节p53表达可能是MCLR促癌过程的重要机制。

    ABSTRACT: Objective To explore the molecular mechanism of microcystinLR(MCLR) inducing of liver tumor. Methods Acording two stagemediumterm theory to set up the animal model, and biochemistry technique to detect the carcinogenesis of the model.RTPCR technique and image analysis were used to study the mRNA expression of the p53 and p16 in the course of inducing tumor. Results (1)MCLR can enhance the positive reaction of γGT. (2)MCLR increased the expression of p53, and have not effect on the expression of p16. Conclusion (1)MC can induced liver tumor formation strongly. (2)The high expression of p53 may play an important role in the carcinogenesis of MCLR in liver. ......
