Abstract
AIM: To investigate the changes of cellular Fas associated death domain-like IL-1β-converting enzyme-inhibitory protein (c-FLIP) expression after the inhibition of protein kinase B (PKB/Akt) activation.
METHODS: Human colon cancer cell line HT-29 was treated with 10, 20 and 40 nmol/L wortmannin (a specific inhibitor for Akt activation) for 0, 3, 6, 12 and 24 hours. MTT assay was used to detect the proliferation of HT-29 cells. After HT-29 cells were treated with 40 nmol/L wortmannin, the levels of phosphorylated Akt (pAkt) and c-FLIP protein were determined by Western blot, and the transcription of c-FLIP gene was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) at the 0, 3rd, 6th, 12th and 24th hour ......
