    Technique modification in establishing rat model of partial liver transplantation with hepatic rearterialization

    Abstract

    AIM:
To develop and modify a rat model of partial liver transplantation with hepatic rearterialization.

    METHODS: Two-cuff technique, described by Kamada and Howden, was applied with some modification and hepatic rearterialization between the donor’s celiac artery and the recipient’s right renal artery was performed in Sprague Dawley rats. The graft reduction was performed ex situ by resecting the left lateral lobe, the left portion of the median lobe, and the caudate lobes.

    RESULTS: Of the 48 rats received liver transplantation, 5 cases died during operation, and the warm ischemia time for the donor liver was 1.0 ± 0.5 minutes. The 1-week survival rate of the recipients was 85.4%.

    CONCLUSION: The modification used in this study provides a more practical and stable experimental model.

    摘要

    目的: 建立大鼠部分肝移植动脉化模型的技术改进. 

    方法:采用改良二袖套法建立大鼠部分肝移植模型, 并进行供体的腹腔动脉与受体的右肾动脉端端吻合. 供肝予以切除尾叶、肝左叶及肝中叶的左半. 

    结果:48例正式实验中, 有5例于术中死亡, 供肝热缺血时间为1.0±0.5 min. 大鼠部分肝移植术后1 wk存活率为85.4%.

    结论: 通过技术改进, 提高了模型建立的稳定性.

    0 引言

    临床上肝移植已成为治疗终末期肝病和先天性肝胆疾病的有效治疗手段[1-3], 活体部分肝移植也由于临床上供肝来源匮乏的原因而得到关注和不断发展, 与活体部分肝移植相关的重要研究是建立稳定的动物模型. 

    1 材料和方法

    1.1 材料♂SD大鼠, 体质量260-280 g, 购于江苏省动物实验中心, 术前禁食20 h, 不禁水, 术前称体质量, 使每对供、受体大鼠体质量相差≤10 g. 手术显微镜(镇江光学仪器厂生产)下清洁手术, 胆总管支架管采用硬膜外导管制成, 长约5.0 mm, 外径0.8 mm. 肝下下腔静脉及门静脉袖套管采用输液皮条拉制而成, 直径分别为2.4-2.6 mm和1.8-2.0 mm, 长约3.0 mm, 袖套管外壁刻烙痕.

    1.2 方法1.2.1 供体手术 盐酸氯胺酮ip麻醉(100 mg/kg), 仰卧位, 十字切口进腹, 离断拉状韧带、左三角韧带及肝胃韧带, 结扎左膈下静脉以及左肝至食管的交通血管, 游离肝下下腔静脉至左肾静脉平面, 游离右肾静脉并用9-0丝线紧贴腔静脉将其单独结扎切断, 游离右肾动脉并结扎, 结扎右肾上腺静脉, 离断肝右后侧所有韧带 ......
