重组人生长激素对肠缺血再灌注大鼠肠壁IgA浆细胞凋亡及P53基因表达影响的实验研究

【摘要】目的:探讨肠缺血再灌注损伤时应用重组人生长激素(rhGH)时肠壁IgA浆细胞的凋亡及P53基因的表达的影响。方法:将60只Wistar大鼠雌雄各半随机分为实验组和对照组,按实验天数(2、4、6d)又各分为3组,每组10只,制造肠缺血再灌注模型,实验组给予rhGH[每日1.33U(kg·d)],观察回肠末端黏膜固有层内IgA浆细胞的凋亡及其P53的表达情况。结果:(1)实验组IgA浆细胞的凋亡在2、4、6天均低于对照组(P<0.05);实验组与对照组第6天IgA浆细胞的凋亡均显著高于第2、4天(P<0.01);(2)实验组IgA浆细胞中凋亡基因P53的表达在第2天和第6天均低于对照组(P<0.05);(3)P53表达与IgA浆细胞的凋亡呈正相关(r=0.812,P<0.05)。结论:rhGH通过抑制IgA浆细胞的凋亡而调节肠免疫屏障,其机制可能是与抑制凋亡基因P53的表达相关。【关键词】促生长素类,重组·肠·再灌注损伤·免疫球蛋白A·浆细胞·基因,P53·大鼠,Wistar

【ABSTACT】Objective:To explore the effect of rhGH on apoptosis and the expression of P53 of IgA plasmocytes of intestinal wall after intestinal ischemiareperfusion injury and the correlation between apoptosis and the expression of P53.Methods:60 Wistar rats were divided into two groups randomly:experiment group(1.33U/kg rhGH was given q.d) and control group.Each group was divided into 3 subgroups(2,4,6days,respectively).Apoptosis and the expression of P53 in IgA plasmocytes of ileal mucosal were observed.Results:(1)Apoptosis of IgA plasmocytes in experiment group was lower than that in control group on 2nd,4th,6th day and had statistical significance(P<0.05);Apoptosis on 6th day was higher than that on 2nd and 4th day in both two groups(P<0.01).(2)The expression of P53 on 2ndand 6thday in experiment group was lower than that in control group ans had statistical significance(P<0.05);(3)The experssion of P53 was correlated positively with apoptosis of IgA plasmocytes(r=0.812,P<0.05).Conclusion:rhGH can inhibit apoptosis of IgA plasmocytes and accommodate the immunologic barrier of intestinal.It come into effect probably by inhibiting the expression of P53. ......
