超声心动图评估静脉注射骨髓间充质干细胞对SD大鼠心肌梗死的治疗效果

[摘要] 目的 以超声心动图评估骨髓间充质干细胞(MSC)静脉注射治疗SD大鼠急性心肌梗死模型的疗效。 方法 结扎SD大鼠冠状动脉左前降支，建立心肌梗死模型，并将模型鼠随机分成盐水对照组(组1)及MSC治疗组(组3)，组2为假手术组(开胸但不结扎冠状动脉)。组1及组3每只鼠分别于再灌注后经尾静脉注射生理盐水1 ml及MSC 1 ml(2.5×106个)，组2不予治疗。4周后对三组鼠行超声心动图检查。统计学处理应用SPSS 11统计软件，所有数据以均数±标准差表示，组间均数比较采用One Way ANOVA分析的LSD法。P<0.05为差异有显著性。 结果 各组间参数均值比较，除组3舒张期左室前壁厚度(AWTed)较组1厚[(0.88±0.16)mm vs (0.68±0.08)mm;P<0.05]外,其余参数两组间差异均无显著性(P>0.05)。免疫组化显示坏死心肌内可见少量移植的MSC向心肌细胞(aactinin染色阳性)及血管内皮细胞(vWF染色阳性)分化。结论 经尾静脉给心肌梗死模型鼠注射MSC，可增加梗死区的室壁厚度，但尚缺乏有效改善心功能的证据。[关键词] 骨髓间充质干细胞；SD大鼠；急性心肌梗死模型；超声心动图
Curative effect of intravenously transplanted mesenchymal stem cells to acute myocardial infarction evaluated by echocardiography

[Abstract] Objective To evaluate the curative effect of intravenously transplanted mesenchymal stem cells(MSC) to acute myocardial infarction(AMI) by echocardiography. Methods SpragueDawley(SD) rats were anesthetized and left anterior descending(LAD) coronary artery was ligated for 1 hour followed by reperfusion.At 24 h after ligation,the rats were randomly divided into two groups to receive:1 ml saline(group 1) or 2.5×106 MSC suspended in 1 ml saline(group 3) by intravenous injection.A thoracotomy was performed but LAD was not ligated in the sham operation group(group 2).Four weeks after the operation,the animals were anesthetized and examined by echocardiography.Then the rats were killed and stained with antibodies to αactinin and vWF. Results In group 3,anterior wall enddiastolic thickness(AWTed) was significantly higher than that of group 1(0.88 mm±0.16 mm vs 0.68 mm±0.08 mm,P<0.05);where the anterior wall amplitude(AWA),interventricular septum enddiastolic thickness(IVSTed) ......
