合成鱼腥草素对小鼠免疫功能的影响

沈阳药科大学学报990315 摘 要 对于脾切除致免疫功能低下小鼠,合成鱼腥草素(HOU)60 mg/kg,120 mg/kg灌胃给药均能增强其腹腔巨噬细胞的吞噬功能(P<0.001);迟发型超敏反应强度(P<0.001);明显提高血清溶血素水平(P<0.05;P<0.001);外周血淋巴细胞ANAE阳性百分率(P<0.001);HOU60 mg/kg还能明显提高脾切小鼠腹股沟淋巴结中淋巴细胞个数(P<0.01),表明HOU对脾切除致免疫功能低下小鼠的特异性、非特异性免疫功能均有明显增强作用.

Abstract For the immunodeficiency mice following splenectomy,houttuyninum ig 60 mg/kg,120 mg/kg strikingly strengthened the phagocytosing function of the peritoneal macrophage system (P<0.001),delayed the type hypersensitivity (DTH)reaction (P<0.001);significantly enhanced the level of serumhemolysin(P<0.05;P<0.01),and ANAE(+) cell percentage of peripheral blood lymphocytes(P<0.001)furthermore,60 mg/kg remarkably raised the amount of lymphocytes of groin lymph node(P<0.05).These experimental results of hottnyninum were suggested that houttuyninum could obviously strengthen the nonspecific and sepecific immune function in splenectomized mice.

合成鱼腥草素(HOU)为中药鱼腥草的有效成分鱼腥草素的亚硫酸钠加成物,具有抑菌、抗炎等方面的药理作用,本室在进行抗炎研究中发现,其对免疫功能低下小鼠具有较好的调节作用.脾为机体最大的免疫器官,脾切除势必造成免疫功能的紊乱.本实验以HOU为供试药物,利用脾切除造成免疫功能低下小鼠模型,对HOU免疫调节机制进行了研究.
1 实验方法
1.1 脾切除方法
用乙醚麻醉小鼠,小鼠俯卧位,左背后部肋缘下切口长约0.5 cm,将脾拉出切口,结扎脾蒂血管,去脾,缝合肌层和皮肤,假手术组同样方法只是不去脾,缝合.术后常规饲养2周,第2周给药.
1.2 分组及给药方法

实验分5组进行,假手术组,脾切除模型组,HOU低剂量组,HOU高剂量组,阳性对照药组.HOU低剂量组,HOU高剂量组分别给予HOU60,120 mg/kg,连续7天,假手术组,脾切除模型组给予等容量的1% CMC,阳性对照药组在最后3天给予盐酸左旋咪唑18 mg/kg,末次给药1 h,按文献方法〔1~6〕进行各种药理指标的测定.
腹腔巨噬细胞的吞噬鸡红细胞能力测定〔1〕,DNCB诱导的迟发型超敏反应强度测定〔2〕,外周血淋巴细胞ANAE阳性百分率测定〔3,4〕,血清溶血素测定〔5,6〕,腹股沟淋巴结中淋巴细胞计数〔6〕见文献.
2 实验结果
2.1 HOU对腹腔巨噬细胞吞噬鸡红细胞能力的影响

如表1所示,脾切除后小鼠腹腔巨噬细胞吞噬鸡红细胞的功能明显降低,HOU60,120 mg/kg均能明显增强脾切除致兔疫功能低下小鼠腹腔巨噬细胞的吞噬功能.

2.2 HOU对DNCB诱导的迟发型超敏反应强度的影响

2.4 HOU对血清溶血素的影响

2.5 HOU对腹沟淋巴结中淋巴细胞计数的影响
