    【摘要】目的:探讨博安霉素(博莱霉素A6)抗人鼻咽癌作用的机理。方法:应用荧光显微镜、流式细胞仪及琼脂糖凝胶电泳等方法检测博安霉素诱导人鼻咽癌SUNE-1细胞凋亡的作用。结果:当博安霉素浓度为50μg/ml,作用于SUNE-1细胞10小时,荧光显微镜下SUNE-1细胞出现明显的凋亡形态学改变;流式细胞仪出现亚G1峰,凋亡率为46.3%;琼脂糖凝胶电泳出现典型的DNA梯状条带。结论:博安霉素具有体外诱导鼻咽癌细胞凋亡的作用,而诱导凋亡为其抗肿瘤作用的机制之一。

    【Abstract】 Objective: To investigate the antitumor mechanisms of boanmycin (bleomycin A6) on human nasopharyngeal cancer. Method: Apoptosis induced by boanmycin in human nasopharyngeal cancer SUNE-1 cells was tested by fluorescent microscope, flowcytometry and DNA gel electro phoresis. Result: When incubated with boanmycin (50 μ g/ml) for 10 hours, SUNE-1 cells showed morphological changes associated with the characters of apoptosis under fluorescent microscope. In flowcytometry analysis sub-G1 peak appeared in DNA frequency distribution histograms. Typical DNA ladder was found during gel electrophoresis. Conclusion: Boanmycin induces apoptosis in SUNE-1 cells in vitro, which may be one of its antitumor mechanisms of action against nasopharyngeal cancer. ......
