    摘 要:目的:用改进的TRAP法检测肺癌的端粒酶活性,探讨端粒酶活性与肺癌的关系。方法:用培养瓶替代液氮瓶收集标本,采用TRAP改进方法,内含36bp质控模板预防假阴性,增加引物长度避免二聚体形成,且使反应一步完成,并用银染法显示检测结果,对45例病理确诊的肺癌手术标本及一株肺癌细胞株的端粒酶活性进行检测。结果:38例肺癌标本端粒酶活性呈阳性,阳性率为84%(38/45),一株肺癌A549细胞株也呈阳性。结论:经改进后的TRAP法检测端粒酶活性,更加准确方便;标本采集处理方法简单有效,易于推广应用;端粒酶活性检测将有助于肺癌的诊断。

    Detection of telomerase activity in lung cancer by modified method

    Abstract:Objective: To determine telomerase activity in lung cancer utilizing modified TRAP method and to reveal the relationship between telomerase activity and lung cancer. Methods: Forty five tumor samples pathologically confirmed lung cancer were collected in culture flasks.The telomerase activity was detected with modified TRAP(telomeric repeat amplification protocol) ,which added a 36bp quality control to prevent false negative , extended the primers length to avoid producing primer diamer and applied silver staine to show the result. Results: Telomerase activity was positive in 38 cases of lung cancer,the positive rate of telomerase activity was 84% . Telomerase activity in a lung cancer cell line A549 was also positive. Conclusion: Modified TRAP for determination of telomerase activity in tumor specimen was easier and more accurate.The preparation of sample was more simple and efficiency,and easy to apply. Thus,detection of telomerase activity by modified TRAP may be useful for the diagnosis of lung cancer. ......
