EMS患者异位和在位内膜组织ROCK1及HOXA10的表达及其意义(1)

     【摘要】 目的 探討子宫内膜异位症(EMS)患者异位和在位内膜组织RhoA/Rho相关激酶1(ROCK1)及同源盒A10(HOXA10)的表达及其意义。方法 收集140例EMS患者的异位和在位内膜组织进行ROCK1及HOXA10表达的免疫组织化学染色(免疫组化)与蛋白免疫印迹法检测,分析患者的临床特征与上述2种蛋白表达水平之间的关系。结果 EMS患者异位内膜组织的 ROCK1表达阳性率为77.9%,高于在位内膜组织的21.4%,异位内膜组织的HOXA10表达阳性率为28.6%,低于在位内膜组织的87.9%(P均< 0.05)。蛋白免疫印迹法显示,异位内膜组织的ROCK1相对表达强度高于在位内膜组织、HOXA10 低于在位内膜组织 (P均< 0.05)。异位内膜组织的ROCK1、HOXA10表达阳性率与EMS患者的年龄、病程、异位内膜分期均无关(P均> 0.05),而与EMS临床分期有关(P < 0.05)。ROCK1与HOXA10表达阳性率有关(P < 0.05)。结论 EMS患者的异位和在位内膜组织中ROCK1、HOXA10表达存在显著差异,推测HOXA10可负调控ROCK1信号通路,从而调节EMS的发生与发展。

    【Abstract】 Objective To investigate the expression and significance of ROCK1 and HOXA10 in ectopic and eutopic endometrium of patients diagnosed with endometriosis (EMS). Methods One hundred and forty patients with EMS admitted to our hospital were enrolled in this study. The ectopic and eutopic endometrial tissues of 140 patients were collected for ROCK1 and HOXA10 expression detected by immunohistochemical staining and Western blot. The relationship between the clinical characteristics and the expression levels of ROCK1 and HOXA10 proteins was analyzed. Results In the ectopic endometrium, the positive rate of ROCK1 was 77.9%, significantly higher than 21.4% in the eutopic endometrium (P < 0.05), whereas the positive rate of HOXA10 was 28.6%, considerably lower than 87.9% in the eutopic endometrium (P < 0.05). Western blot revealed that the relative expression of ROCK1 in the ectopic endometrium was significantly higher, whereas the relative expression of HOXA10 was remarkably lower than those in the eutopic endometrium (both P < 0.05). In the ectopic endometrium, the positive rates of ROCK1 and HOXA10 were not significantly correlated with the age, course and staging of ectopic endometrium (all P > 0.05), whereas were significantly associated with the clinical staging of EMS (P < 0.05). Association analysis of categorical variables showed that the positive rate of ROCK1 was correlated with that of HOXA10 (P < 0.05). Conclusions The expression levels of ROCK1 and HOXA10 significantly differ between the ectopic and eutopic endometrial tissues of EMS patients.
