     (广西医科大学第一附属医院心内科广西南宁530021)【基金项目】博士启动基金编码:308032【摘要】目的 观察阿托伐他汀预处理对损伤后大鼠心肌细胞MMP-2表达和细胞凋亡的影响。方法 体外培养大鼠心肌细胞随机分成4组:对照组; 缺氧-复氧组(HR组);HR组+0.1μmol/L阿托伐他汀;HR组+1μmol/L阿托伐他汀;用ELISA试剂盒检测细胞上清液MMP-2各组的表达,流式细胞仪分析细胞的凋亡率(AR)。结果 ①与对照组比较,HR组MMP-2表达和细胞凋亡率明显上升(P<0.05)②与HR组比较,低剂量阿托伐他汀药物组MMP-2表达和细胞凋亡率均下调,但差异没有统计学意义(P >0.05)③高剂量阿托伐他汀药物组MMP-2分泌量和细胞凋亡率低于低剂量组,差异有统计学意义(P<0.05)。结论 阿托伐他汀能够降低损伤心肌细胞MMP-2的分泌和细胞凋亡。【关键词】阿托伐他汀;MMP-2;缺氧复氧损伤;细胞凋亡 【中国分类号】R722.1 【文献标识码】A【文章编号】1004-5511(2012)04-0040-02 【Abstract】Objective To observe the effect of atorvastation preconditioning on the expressions of MMP-2 and Cardiomyocyte Apoptosis after hypoxia/rexygen injury in Rats. Methods To culture cardiomyocyte cells and divided into four groups randomly.Group C (n=6);the hypoxia/reoxygen(H/R)group;HR+0.1μmol/L atorvastation group;HR+μmol/L atorvastation. MMP-2 and Cardiomyocyte Apoptosis were detected. Results The level of MMP-2 and Cardiomyocyte Apoptosis of HR group were significantly higher than C group(P<0.05);The leve of MMP-2 and Cardiomyocyte Apoptosis more Cardiomyocyte were lower than the less group (p<0.05).
