双酚与氯化镉对乳腺癌细胞MCF-7增殖作用的研究(1)

     【摘要】目的 用环双酚A(BisphenolA,BPA)和氯化镉单独染毒和联合染毒乳腺癌MCF-7细胞进行研究,评价双酚A和氯化镉的联合作用类型及环双酚A活性。方法 MTT法检测BPA和氯化镉对MCF-7细胞增殖效应的单独作用及两者间联合作用的剂量效应关系;观察不同剂量组的BPA和氯化镉分别作用于MCF-7细胞时,对细胞增殖的影响。实验设立了溶剂对照组,同时设雌二醇为阳性对照组。设BPA和氯化镉浓度均为1.0×10-5、1.0×10-6、1.0×10-7 mol/L时,观察两者对MCF-7细胞的联合作用。结果 MTT结果显示,BPA浓度为1.0×10-9~1.0×10-4 mol/L范围、氯化镉浓度在1.0×10-8~1.0×10-5 mol/L范围单独作用时均能促进细胞增殖,并呈倒U型剂量效应曲线,其中BPA在1.0×10-6mol/L,氯化镉在1.0×10-6mol/L浓度下促增殖效应均达到最大,其相对增殖效应分别为2.091和1.3528,与对照组比较差异均具有统计学意义(P<0.05)。在BPA与氯化镉均为1.0×10-5、1.0×10-6、1.0×10-7 mol/L浓度联合染毒时,对MCF-7细胞的增殖作用较单独作用显著降低,表现出明显的拮抗作用。结论 BPA、氯化镉对MCF-7细胞均有增殖的效应,在一定浓度范围内呈现低浓度促进,高浓度抑制的作用;两者之间的联合作用为拮抗作用。

    【Abstract】Objective study on the effect of exposure separately and jointly with the bisphenol A and cadmium chloride by breast cancer MCF-7 cells, so as to evaluate environmental estrogenic activity and combined effect type of bisphenol A and cadmium chloride. Method Dose-effect relationship of the effect bisphenol A and cadmium chloride on was detected separately and jointly by MTT to emulate; observed the effect of bisphenol A and cadmium chloride with different concentration on MCF-7 cells multiplication. Control group was established, the effect of bisphenol A and cadmium chloride with 1.0×10-5、1.0×10-6、1.0×10-7mol/L on MCF-7 cells was analyzed Result the results showed that alone group of bisphenol A with range of concentration 1.0×10-9 to 1.0×10-4 mol/L and cadmium chloride with 1.0×10-8 to 1.0×10-5 mol/L respective can promote cell proliferation. To take on U-shaped dose-response curve, the effect of cell proliferation was strong with bisphenol A of 1.0×10-6mol/L and cadmium chloride of 1.0×10-6mol/L, the multiplication effective were 2.091 and 1.3528,There were statistically significant compared to control group ......
