     【摘要】目的:研究芹菜素(apigenin, AP)对糖尿病模型小鼠氧化应激和单核细胞趋化蛋白-1 的影响,并初步探讨其降血糖的作用机制。方法:复制链脲佐菌素(STZ)致糖尿病小鼠模型。将50只昆明鼠随机分为5组,分别是正常对照组、模型组、AP 低(5 mg•kg-1)、中(10 mg•kg-1)和高(20 mg•kg-1)剂量治疗组,治疗组每日腹腔注射给药,连续30 d。采用血糖仪检测空腹血糖;ELISA 法检测血清中超氧化物歧化酶(SOD) 活性和丙二醛(MDA)含量;RT-PCR 法检测胰腺单核细胞趋化蛋白-1(MCP-1) mRNA 表达。结果:AP 可显著降低糖尿病模型小鼠血糖,提高SOD 活性,减少MDA 含量,抑制胰腺MCP-1mRNA 表达(P < 0.05, P < 0.01),并具有药物剂量依赖性。结论:AP 对STZ 引起的高血糖有很好的降血糖作用,其作用机制可能与其降低氧化应激水平和抑制MCP-1 的表达相关。

    The Effect of Apigenin on Oxidative Stress and Monocyte Chemoattractant Protein 1 in Diabetic Mice

    【Abstract】Objectiv: To study the effect of apigenin on oxidative stress and monocyte chemoattractant protein 1 in diabetic mice, and explore its mechanism of hypoglycemosis. METHODS: Ddiabetic mice model was established by streptozotocin(STZ). Fifty kunming mice were divided into five groups randomly, including normal control group, model control group, AP low dose(5 mg•kg-1) treatment group, AP middle dose(10 mg•kg-1) treatment group and AP high dose(20 mg•kg-1) treatment group. The AP treatment groups were administrated AP i.p. dailyfor thirty days continuously. Fasting blood glucose was tested by glucose meter; the activity of superoxide dismutase(SOD) and the level of malondialdehyde(MDA) in blood serum were tested by ELISA; the expression of monocyte chemoattractant protein 1(MCP-1) mRNA in pancreas was tested by RT-PCR. RESULTS: AP can decrease the content of blood glucose, increase the activity of SOD, lower the level of MDA, inhibit the expression of MCP-1 mRNA in a dose-dependent manner in diabetic mice induced by STZ significantly(P < 0.05, P < 0.01). CONCLUSION: AP has a good hypoglycemic effect on diabetic mice induced by STZ, and its mechanism may be related to lower the level of oxidative stress and inhibit the expression of MCP-1.
