【摘要】
目的：通过动物体内实验研究证实rhuCD40L对人结肠癌的治疗作用，从而进一步探讨其抗肿瘤机制。
方法：将皮下接种HT-29人结肠癌细胞后建立人结肠癌模型的20只裸鼠随机分为二组，治疗组以rhuCD40L作皮下注射；对照组以生理盐水作皮下注射，定期观察裸鼠的生活状况并测量其体重，治疗5周后处死裸鼠并取瘤，测量两组瘤大小及瘤重，并比较结果。
结果：20只裸鼠全部成瘤，实验过程中所有裸鼠生活状况均良好，无一裸鼠死亡；对照组与治疗组之间的瘤大小及瘤重存在差异，差异有统计学意义(P<0.05)。
结论：rhuCD40L对人结肠癌有一定的治疗作用。

The therapeutic action of rhuCD40L to nude mice with person colon cancer hypodermic transplant lump

【Abstract】Purpose: To confirms the therapeutic action of rhuCD40L through the animal in vivo experimental study to the person colon cancer , thus further discusses its anti-tumor mechanism.Methods: After hypodermic vaccinating the HT-29 person colon cancer cell, We established the person colon cancer model of 20 nude mice,then divided them into two groups stochastically, the treatment group made the hypodermic injection by rhuCD40L, the control group made the hypodermic injection by the physiological saline, observed their living condition regularly and surveyed their body weight, treating 5 weeks later executed the nude mice and took the lumps, surveyed the sizes and the weight of them, and compared the result.Result: 20 nude mice carried the lump completely, in the experiment process all nude mice were good living condition, no one nude mice died. There are difference between the control group and the treatment group's lump size and the lump weight , the difference has statistics significance (P<0.05).Conclusion: RhuCD40L has certain therapeutic action to the person colonic cancer.

CD40L对表达CD40的肿瘤细胞具有广泛的生长调节作用。大量研究表明，CD40L与表达CD40的肿瘤的相互作用可以直接抑制多种肿瘤的生长。结肠癌是胃肠道中常见的恶性肿瘤，在我国近20年来尤其在大城市，发病率和病死率有逐年上升的趋势。目前治疗结肠癌仍以手术切除为首选治疗方法，但由于术后局部复发和转移率较高，术后5年生存率仍较低。当前需要寻求一种更好的辅助治疗手段以提高结肠癌患者生存率。因此，本实验以裸鼠为载体，选用人结肠癌HT-29细胞株建立人结肠癌裸鼠皮下移植模型，旨在研究rhuCD40L的抗肿瘤作用。
1材料与方法
1.1 细胞:HT-29人结肠癌细胞株，由中国科学院上海细胞库提供。
1.2 实验动物: 20只SPF级裸小鼠，4～5周龄，体重16～20克，均为雌性，由中国科学院上海实验动物中心提供。均饲养于苏州大学SPF级实验动物中心。
