基于网络药理学的安肠愈疡汤治疗溃疡性结肠炎作用机制研究(1)

     摘要:目的 基于网络药理学探讨安肠愈疡汤治疗溃疡性结肠炎(ulcerative colitis,UC)的作用机制。方法 通过中医药整合药理学研究平台构建安肠愈疡汤的潜在靶标与UC疾病靶标之间的相互作用网络。基于GO数据库和KEGG通路数据库进行基因功能和通路富集,采用Cytoscape3.6.1软件构建靶标-信号通路网络、药物活性成分-靶标网络,采用荧光实时定量PCR对网络药理学主要分析结果进行验证。结果 分析结果表明,安肠愈疡汤靶点主要关联UC炎症因子及免疫调控,糖类、脂质和氨基酸代谢,能量代谢,神经-内分泌系统调节等;体外实验提示,安肠愈疡汤参与调控GCK、CROT介导的糖脂代谢,GNB1介导的炎症通路,以及ATP1A1、COX7C介导的能量代谢、氧化磷酸化、线粒体电子呼吸链传递等通路。结论 安肠愈疡汤治疗UC具有多成分、多靶点的特点,可为进一步研究其作用機制提供依据。

Study on Mechanism of AnchangYuyang Decoction in Treating Ulcerative ColitisBased on Network Pharmacology

    Abstract: Objective To explore the mechanism of Anchang Yuyang Decoction in the treatment of ulcerative colitis (UC) based on network pharmacology. Methods The interaction network between UC targets and potential targets of Anchang Yuyang Decoction was constructed through Integrative Pharmacology-based Research Platform of TCM. Gene function and pathway enrichment were conducted based on GO database and KEGG pathway database. Cytoscape3.6.1 was used to construct target-signal pathway network and pharmaceutically active component-target network. The main results of network pharmacology were verified by real-time quantitative PCR. Results The results showed that the targets of Anchang Yuyang Decoction were mainly associated with inflammatory chemokines and immune system regulation in UC, carbohydrates, lipids and amino acid metabolism, energy metabolism, neuro-endocrine system regulation, etc. Vitro experiments indicated that Anchang Yuyang Decoction was involved in the regulation of glucose and lipid metabolism mediated by GCK and CROT, inflammatory pathway mediated by GNB1, as well as energy metabolism, oxidative phosphorylation, mitochondrial electron respiratory chain transmission mediated by ATP1A1 and COX7C. Conclusion AnchangYuyang Decoction applied for treating UC has the advantages of multi-components and targets, which can provide references for further research on mechanism.

    溃疡性结肠炎(ulcerative colitis,UC)是一种以结肠和直肠黏膜弥漫性炎症为特征的慢性非特异性炎症性肠病。本病临床症状以发作、缓解交替出现为特点,易复发,且部分重症患者病情重、变化快,药物治疗效果不佳。目前,现代医学对UC病因及发病机制未完全阐明,尚无针对其病因的特效药物。中医通过辨病与辨证相结合,抓住UC病机特点,随证加减用药,可显著改善UC患者生活质量,维持疾病临床缓解,防治并发症,并降低疾病复发率[1], http://www.100md.com(樊静娜)
