神经生长因子受体p75NTR在喉癌Hep2细胞系中的表达(1)

     摘 要:目的 初步探索神经生长因子受体在喉癌Hep2细胞系的表达情况并研究相关的细胞生物学特性。方法 应用免疫细胞化学及免疫荧光染色法,检测p75NTR在喉癌Hep2细胞系中的表达;应用流式细胞仪检测人喉癌Hep2细胞系中呈p75NTR高表达及CD133高表达细胞的分布比例情况。结果 喉癌Hep2细胞系内小部分肿瘤细胞显示免疫染色强阳性,p75NTR强阳性显色的细胞部分处于旺盛的增殖分裂状态;大部分呈CD133强阳性的Hep2细胞也呈p75NTR强阳性。结论 p75NTR高表达于喉癌Hep2细胞系中具有较强增殖活力的细胞,p75NTR信号通路对支持或促进喉癌细胞增殖具有潜在作用。p75NTR高表达的喉癌Hep2细胞与肿瘤干细胞之间的关系密切,有待进一步深入研究。

    Expression of Nerve Growth Factor Receptor p75NTR in Hep2 Cell Line of Laryngeal Squamous Cell Carcinoma

    Abstract:Objective To investigate the expression of nerve growth factor receptor in laryngeal squamous cell carcinoma Hep2 cell line and to study the related cellular biological characteristics.Methods Immunocytochemistry and immunofluorescence staining were used to detect the expression of p75NTR in Hep2 cell line of laryngeal squamous cell carcinoma,and the distribution of p75NTR and CD133 in Hep2 cell line of laryngeal squamous cell carcinoma was detected by flow cytometry.Results A small number of tumor cells in the Hep2 cell line of laryngeal squamous cell carcinoma showed strong immunostaining,and the cells with strong positive coloration of p75NTR were in a proliferative and dividing state;most of the Hep2 cells positive for CD133 were also strongly positive for p75NTR.Conclusion p75NTR is highly expressed in the Hep2 cell line of laryngeal squamous cell carcinoma and has strong proliferative activity.The p75NTR signaling pathway has a potential role in supporting or promoting the proliferation of laryngeal squamous cell carcinoma cells.The relationship between p75NTR-expressing laryngeal squamous cell carcinoma Hep2 cells and tumor stem cells is closely related and needs further study.

    喉癌(laryngeal squamous cell carcinoma,LSCC)是头颈外科临床常见恶性肿瘤,喉癌通常可以损害患者的发音、呼吸、吞咽等功能,同时又具有破坏力强、易复发、易转移等特点,临床上很多患者难以治愈甚至危及生命。研究喉癌发病及恶性进展的相关机制,有助于探索治愈喉癌的有效途径。近年来肿瘤干细胞理论逐渐成熟,众多学者认为肿瘤组织内部包含少量拥有自我更新能力、分化能力及放化疗抵抗能力的特殊细胞群,与肿瘤的治疗后复发密切相关[1]。神经生长因子受体p75NTR(p75 neurotrophin receptor,p75NTR)是肿瘤坏死因子受体(tumor necrosis factor receptor,TNFR)超家族成员之一,广泛分布于人体各组织系统及肿瘤细胞中[2]。除中枢及外周神经系统外,p75NTR蛋白也广泛地表达于上皮、间叶及淋巴造血组织等非神经组织系统。研究证实p75NTR在不同组织细胞类型、不同病理生理环境中执行的功能具有多样性,能够调节细胞的存活、增殖、分化、凋亡及创伤修复等效应[3]。近年来p75NTR在肿瘤细胞中的存在及作用引起广泛关注,目前国内外关于p75NTR在喉癌细胞系中的表达及功能文献少有报道,我们采用免疫细胞化学及流式细胞仪等方法研究p75NTR在喉癌Hep2細胞系中的表达规律和潜在作用,为进一步研究喉癌的综合治疗提供理论依据。, 百拇医药(申宇鹏 李晓明 靳海峰 路秀英)
