基于网络药理学探究中药川芎治疗高血压的药理作用机制(1)

     【摘 要】 目的:采用網络药理学的研究方法,探讨活血化瘀中药川芎治疗高血压的药理作用机制。方法:通过中药系统药理学数据库(TCMSP)检索并筛选川芎的有效成分及其作用靶点,通过GeneCards、OMIM等数据库检索与高血压相关基因,利用Cytoscape构建并合并活性成分-靶点网络和疾病-靶点网络,通过网络拓扑分析筛选核心靶点,并采用DAVID数据库进行关键靶点的GO分析和KEGG通路富集分析,探讨其治疗高血压的分子机制。结果:通过数据库筛选和文献查阅共得到川芎有效成分23个,得到川芎与高血压的交集靶点为41个;将这些靶点进行PPI网络分析,得到degree较大的14个靶点蛋白,表明它们是川芎治疗高血压的主要作用靶点;并且发现这些关键靶点与15条信号通路相关,分别是HIF-1信号通路、钙信号通路、VEGF信号通路、PI3K-Akt信号通路、cGMP-PKG信号通路、雌激素通路等。结论:本研究从网络药理学的角度初步揭示了川芎治疗高血压的分子机制,为其进一步的药效物质基础分析和分子机制研究提供一定依据。

    Study on Mechanism for Treating Hypertension of Ligusticum chuanxiong Hort. Based on Network Pharmacology

    Abstract:Objective Use the network pharmacology research method to explore the pharmacological mechanism of Chuanxiong, a traditional Chinese medicine for promoting blood circulation and removing blood stasis. Methods The active constituents of Chuanxiong and their targets were searched and screened by the Chinese Medicine System Pharmacology Database (TCMSP). Genes related to hypertension were searched by GeneCards, OMIM and other databases. Cytoscape was used to construct and combine active component-target network and disease-target network. The core target was screened by network topology analysis, and the DAVID database was used for GO analysis of key targets and KEGG pathway enrichment analysis to explore the treatment of hypertension. The molecular mechanism. Results A total of 23 active constituents of Chuanxiong were obtained through database screening and literature review, and 41 cross-linking targets were obtained. The target sites were analyzed by PPI network to obtain 14 target proteins with a greater degree. They are the main targets for the treatment of hypertension; and these key targets are found to be associated with 15 signaling pathways: HIF-1 signaling pathway, calcium signaling pathway, VEGF signaling pathway, PI3K-Akt signaling pathway, GMP-PKG Signal pathways, estrogen pathways, etc. Conclusion This study reveals the molecular mechanism of Chuanxiong in the treatment of hypertension from the perspective of network pharmacology, and provides a basis for further research and molecular mechanism research.
