针刺足三里对小鼠皮肤光老化的影响(1)

     [摘要]目的:研究针刺足三里对紫外线照射引起的无毛小鼠皮肤光老化模型皮肤组织丙二醛(MDA)含量、超氧化

    物歧化酶(SOD)活力的影响。方法:实验选取2月龄健康昆明种无毛小鼠32只,雌雄各半,体重18~22g。随机分成4组:正常对照组、模型组、针刺组、电针组,每组8只。用紫外线灯管模拟紫外线照射,造成无毛小鼠皮肤光老化,造模同时针刺小鼠足三里,采用生化分析方法测定SOD活性及MDA含量。结果:与对照组比较,模型组无毛小鼠皮肤组织SOD活性明显降低(P<0.01),MDA 含量明显增加(P<0.05);与模型组比较,针刺组和电针组对SOD 活性有提高作用(P<0.05),并可降低MDA 含量(P<0.01)。结论:针刺足三里对紫外线照射引起的无毛小鼠皮肤光老化具有保护作用。其机制可能是:一方面足三里本身具有清除自由基的作用;另一方面可以通过提高SOD活力来达到抗氧化的目的,从而达到抗皮肤光老化的目的。

    Influence to the skin-photoagedemouse caused by acupunctrueing Zusanli

    Abstract:Objective To investigate acusing Zusanli acupoint influence on the content ofMalondialdehyde(MDA) in the skin of hireless photoageing mice as well as the activity of Superoxide Dismutase (SOD) in their skin. Methods Healthy KM hireless mice were divided into four groups randomly and gavaged respectively, that is negative Control Group (A),Model Group (B),needling group(C), electroacupuncture group(D),The mice in B,C,D group were irradiated to simulate skin photoageing by long-term ultraviolet irradiation, at the same time ,the C,D group were acupunctured in Zusanli acupoint. While the changes of the contents of Malondialdehyde,as well as the activity of Superoxide Dismutase were measured by biochemical test method in each group of the experiment. Results Compared with the group A , In model group ,the activity of SOD was decreased(P<0.01) ,while the content of the MDA was increased seriously(P<0.05). Compared with the group B, The contents of MDA in dermal in group C and D had difference with that in group B ,the activity of SOD in dermal in group C and D was increased(P<0.05),while the content of MDA was reduced(P<0.01). Conclusions Acusing Zusanli acupoint has theprotective effect on the skin of the photoageing hairless mice. The mechanism may be that on one hand, Zusanli acupoint could eliminate free radicals itself, on the other hand , by increasing the activity of SOD, it could inhibit the oxidative damages of ultraviolet and skin photoage.

    随着人们生活水平的提高,人们对美的要求也逐渐升温,养颜驻颜已成为众多学者研究的内容。针灸美容越来越受到学者们的认识,针灸美容作为一种常用的美容方法也逐渐得到认可。为了探讨针灸美容的机制,我们以紫外线照射引起的无毛小鼠皮肤光老化模型作为研究对象,进行了本项研究,现将结果报道如下。

    1材料和方法

    1.1材料:由辽宁中医药大学实验动物中心提供健康昆明种小鼠,2个月龄,雌雄各半,体重18~22g。-70℃冰箱、匀浆器、离心机、恒温水浴箱、分光光度计、各种手术用的医疗器械(均由辽宁中医药大学基础医学院分子生物学实验室提供),针刺用针(0.5寸毫针)、韩氏WQl002F型多用电针仪由辽宁中医药大学实验针灸教研室提供。10%水合氯醛,SOD、MDA测定试剂盒(均购于南京建成生物工程研究所)。

    1.2方法

    1.2.1实验动物及分组:将32只无毛小鼠随机分成4组:正常对照组、模型组、针刺组、电针组,每组8只。每组无毛小鼠按性别及组别分笼饲养,其中雄性鼠全部分笼饲养。在自然、相同环境下,常规饲食饮水,无不良因素影响,饲养1周适应实验室环境后开始进行实验。

    1.2.2造模前准备:将实验用及针刺用小鼠称重,然后腹腔注射10%水合氯醛麻醉(0.35ml/100g),待麻醉后用绳将其仰面捆绑在鼠架上,用碘伏消毒足三里部位。

    1.2.3针刺方法:针刺组:以32号0.5寸毫针针刺小鼠膝关节外侧腓骨小头下3.5mm“足三里”穴,直刺3.5mm,平补平泻针法,留针10min,隔日一次,共持续30天。电针组:电压l~3V,疏密波,频率2~20Hz,刺激时间10min,以小鼠肢体或针柄微微颤动为度。隔日一次,共持续30天。, http://www.100md.com
