基质金属蛋白酶-12及其组织抑制物-1在皮肤鳞状细胞癌中的表达及意义(1)

[摘要]目的：检测基质金属蛋白酶-12(MMP-12)及其组织抑制物-1(TIMP-1)在皮肤鳞状细胞癌(SCC)中的表达并探讨其与肿瘤分化、淋巴转移的关系。方法：免疫组化法检测MMP-12及TIMP-1在20例正常皮肤组织和32例鲍温病(BD)、53例SCC患者皮损中的表达。结果：在正常人皮肤和BD患者皮损中，MMP-12不表达，TIMP-1呈弱阳性表达，而两者在皮肤SCC患者皮损中均呈弥漫表达，MMP-12在SCC患者瘤组织中表达阳性率为92.4%，在低分化组或有淋巴结转移组中的表达明显高于高分化组(P＝0.009)或无淋巴结转移组(P＝0.01)；TIMP-1在SCC患者皮损中表达阳性率为84.9%，在高分化组或无淋巴结转移组中的表达明显高于低分化组(P＝0.005)或有淋巴结转移组(P＝0.01)。结论：MMP-12、TIMP-1蛋白的表达可能与SCC的癌分化和转移有关，MMP-12高表达与TIMP-1低表达提示SCC肿瘤细胞可能有较高的侵袭潜能。

Abstract:ObjectiveTo investigate the expression of matrix metalloproteinase-12(MMP-12)and its tissue inhibitor-1(TIMP-1)in cutaneous squamous cell carcinoma(SCC)and the relationship between the expression and tumor differentiation and lymph node metastasis.MethodsImmunohistochemical method was used to detect the expression of MMP-12 and TIMP-1 proteins on the paraffin embedded sections of 53 patients with SCC and 32 patients with BD.ResultsMMP-12 and TIMP-1 proteins were diffusely expressed in the tumor nets and mesenchymal cells around the nets,while in normal epidermis and BD MMP-12 was negative,and TIMP-1 was weakly positive. The expressive rate of MMP-12 was 92.4%,and the expression in poorly differentiated tumous or with lymph node metastasis was significantly higher than that in highly differentiated ones(P＝0.009)or without lymph node metastasis (P＝0.01).
