干扰素对腭裂术后裸露骨面瘢痕成纤维细胞生物学影响的实验研究

     [摘要]目的:探讨干扰素(IFN)对体外培养的腭裂术后裸露骨面瘢痕来源的成纤维细胞生物学作用的影响。方法:制作兔腭裂术后裸露骨面瘢痕动物模型,利用体外细胞培养技术,通过MTT法、流式细胞术等方法观察、分析rhIFNα-2b和rhIFN-γ对腭裂术后瘢痕成纤维细胞生物学行为的影响,数据采用方差分析及q检验处理。结果:与对照组比较,实验组加入干扰素后,腭裂术后瘢痕成纤维细胞的生长趋势及增殖活性及被明显抑制,S-G2-M期细胞百分数明显降低。结论:干扰素对腭裂术后瘢痕成纤维细胞的生长趋势及增殖活性具有明显抑制作用,说明在防治腭裂术后裸露骨面瘢痕的形成中有一定的应用价值。

    Empirical study of interferon affecting on the fibroblasts of the exposed bone surface from cleft palate scar

    Abstract:ObjectiveTo study the effects of interferon to the biological activities of fibroblasts origined from the exposed bone surface from cleft palate scar in vitro cultural. MethodsMaking rabbit cleft palate scar models, in vitro culture,observed by MTT assay and flow cytometry, analysis the effects of interferon to the biological activities of fibroblasts origined from the exposed bone surface of cleft palate scar. Data using analysis of variance and q test treatment. ResultsCompared with the control group,after added interferon, cleft palate fibroblast's growth trends and value-added activity of experimental group was inhibited significantly. Percentage of cells decreased during the S-G2-M stage.ConclusionsInterferon on the growth trends and value-added activity of the cleft palate fibroblast has significant inhibitory effect, indicating that cleft palate in the prevention of scar formation of the exposed bone surface has some value.

    先天性腭裂发病率约占人口出生率的1/700左右,严重影响患者的口腔颌面部器官的形态与功能及心理健康。虽然目前的外科技术已能实现对腭部裂隙的良好修复,有助于语音及吞咽功能的恢复,但大量的临床与基础研究证实,术后患者上颌骨的生长发育继发畸形会变得更为严重,对患者的颌面形态与功能构成严重危害。腭裂术后上颌骨继发生长畸形的病理解剖学基础已经探明是腭裂术后遗留在上颌骨硬腭的裸露骨面瘢痕挛缩所致[1-2]。因此,如何防治腭裂术后裸露骨面瘢痕形成是目前口腔颌面外科领域亟待解决的难题之一。在瘢痕的形成与增生过程中,成纤维细胞是主要的效应细胞,其功能状态是影响瘢痕发生、发展、以及转归的主要因素。干扰素(IFN)具有抗病毒、抗肿瘤、影响细胞增殖分化等多种生物学效应。临床上已经有将干扰素用于防治瘢痕形成的报道,但用干扰素观察腭裂术后裸露骨面瘢痕成纤维细胞生物学行为的影响尚未见报道,本实验通过体外观察和研究干扰素对腭裂术后裸露骨面瘢痕成纤维细胞的生物学作用,可为寻求应用干扰素治疗腭裂术后裸露骨面瘢痕提供实验依据 ......
