硒化壳聚糖促表皮细胞增殖作用的实验研究

     [摘要]目的:研究硒化壳聚糖对体外培养表皮细胞增殖的影响。方法:用不同剂量(25、50、100、200、400mg/L)硒化壳聚糖作用于表皮细胞,电镜观察药物对细胞超微结构的影响; 3H-TdR掺入法、LDH漏出率用于检测药物对细胞增殖及损伤的影响,并以等体积的细胞培养液处理为对照组。结果:各药物浓度作用细胞后,细胞核均有不同程度的增大,核浆比增大,核仁变大,变多,扩张的内质网增多,细胞表面突起增加,线粒体更为丰富,更易见到聚集成团的糖原等现象;硒化壳聚糖可促进表皮细胞对3H-TdR的吸收(P<0.05,P<0.01),降低LDH漏出率(P<0.05,P<0.01)。结论:硒化壳聚糖能促进体外培养表皮细胞增殖。

    The promoting proliferation effect of selenium chitosan on the epidermal cell cultured in vitro

    Abstract: ObjectiveTo study the effects of selenium chitosan on the proliferation of culturedEpidermal cell in vitro.MethodsThe culturedEpidermal cell were treated with selenium chitosan at concentrations of 25、50、100、200、400 mg/Land the equal volume of media as control group.the ultrastructures of Epidermal cell were studied under electron microscope; the effect on cell proliferation was measured by 3H-TdR infiltration and the damage to cell was measured by the leakage rate of LDH.ResultsThe electron microscope showed that in the test group the nucleus endoplasmic reticulum mitochondrion glycogen and the ratio of nucleus to cytoplasm increased or became bigger in a concentration dependence manner. Compared with control group, selenium chitosan could enhance the absorption to3H-TdR (P<0.05, P<0.01)and decrease the leakage rate of LDH (P<0.05, P<0.01).ConclusionSelenium chitosan could promote the proliferation of Epidermal cell cultured in vitro.
