葛根及葛根素对自然衰老小鼠CAT活力及H2O2含量影响的实验研究(1)

[摘要]目的：研究与探讨葛根对小鼠自然衰老的延缓作用。方法：健康4月龄雌性昆明种小鼠15只，作为青年对照组(A)；健康18月龄雌性昆明种小鼠90只，随机分为6组：老年空白对照组(B)、老年阳性对照组(C)、老年葛根中剂量组(D)、老年葛根高剂量组(E)、老年葛根素中剂量组(F)、老年葛根素高剂量组(G)。以灌胃方式给药，以生化分析方法测定过氧化氢酶(CAT)及过氧化氢(H2O2)含量的影响。结果：与青年对照组比较，其余六个老年组CAT的活力下降，H2O2含量显著增高；与老年空白对照组比较，其余五个老年用药组均可提高CAT活性，降低H2O2含量。结论：葛根及葛根素具有延缓小鼠自然衰老的作用。

Effect of radices puerarire and puerarin on activities of CAT and content of H2O2 of naturally senile mice

Abstract: Objective To study and discuss that radices puerarire and puerarin have function on delaying naturally senile mice. Methods Healthy 4 month old mice were 15, as the young control group (A); healthy 18 month old mice were 90 as the elderly group, and then randomly divided into 6 groups:elderly control group (B), positive control group elderly (C),middle puerarin dose group(D), high puerarin dose group(E), middle radices puerarire dose group(F),high radices puerarire dose group(G). Giving medicine by filling stomach. Determined by biochemical analysis of CAT and H2O2 content. Results Compared with the young control group,the remaining mice in each six elderly aged group's the vitality of CAT decreased significantly, H2O2 in serum were significantly higher. Compared with the elderly control group, treatment group of other elderly can improve the vitality of CAT and reduce H2O2 content. Conclusion Radices puerarire and puerarin have function on delaying the naturally senile mice.

葛根为常用中药，是豆科植物野葛或干葛藤的干燥根，干葛藤又可称为粉葛，始载于《神农本草经》，被列为中品，有“山人参”的美称。葛根性平，味甘辛，具有发表解肌、开阳透疹、解热生津等作用。现代研究表明，葛根具有多种药理作用，如抗心律失常、降血糖、抗氧化、抗癌等。本课题通过观察葛根及葛根素对自然衰老小鼠血清中CAT活力及H2O2含量的影响，探讨葛根及葛根素是否具有抗皮肤衰老的作用，为研究和开发延缓衰老的药物提供实验依据。
1 材料和方法
1.1 实验动物及分组方法：健康雌性4月龄昆明种小鼠15只，作为青年对照组(A)；健康雌性18月龄昆明种小鼠90只作为老年组，再随机分成6组，每组15只：老年空白对照组(B)、老年阳性对照组(C)、老年葛根中剂量组(D)、老年葛根高剂量组(E)、老年葛根素中剂量组(F)、老年葛根素高剂量组(G)。
1.2 实验药物及给药方法：实验药物：中药葛根(由辽宁中医药大学附属医院中药药剂科提供)；葛根素(购自成都曼斯特生物科技有限公司)；维生素EC混悬液(购于成大方圆药房，药品批号，国药准字H11022348北京双鹤药业股份有限公司) ......
