三种不同波长激光照射对体外培养的成纤维细胞Ⅰ型和Ⅲ型前胶原mRNA表达水平的影响(1)

     [摘要]目的:对体外培养的小鼠成纤维细胞分别使用3种不同波长(595nm、755nm、1 064nm)的激光照射,观察分析不同波长激光照射对成纤维细胞中I型、Ⅲ型前胶原mRNA表达水平的影响。方法:采用体外培养新生小鼠成纤维细胞,根据激光照射的波长不同通将其分为对照组6只(不采用激光照射),595nm组6只(采用波长为595nm激光照射),755nm组6只(采用波长为755nm激光照射)及1 064nm组6只(采用波长为1 064nm激光照射)。通过荧光定量(RT-PCR)法测定成纤维细胞I型、Ⅲ型前胶原mRNA的表达水平。结果:4组间Ⅰ、Ⅲ型前胶原mRNA的表达水平间差异均有统计学意义(P<0.05);其中1 064nm组的Ⅰ、Ⅲ型前胶原mRNA的相对表达量达到最高,分别为(113.07±2.85)×10-2、(118.72±1.96)×10-2,均显著高于595nm组的[(96.74±1.03)×10-2、(101.51±1.64)×10-2,P<0.01],755nm组的[(100.02±1.86)×10-2、(109.27±1.02)×10-2,P<0.01],及对照组的[(87.14±0.82)×10-2、(100.48±0.73)×10-2,P<0.01];755nm组Ⅰ型前胶原mRNA的相对表达量显著高于对照组(P<0.01),而其Ⅲ型前胶原mRNA的表达水平则显著高于595nm组和对照组(P<0.01);595nm组仅有Ⅰ型前胶原表达水平mRNA显著高于对照组(P<0.01)。结论:激光照射可以诱导成纤维细胞中I型、Ⅲ型前胶原mRNA表达增加。其中,波长为595nm的激光照射仅能促进I型前胶原mRNA表达上调,755nm激光照射后Ⅲ型前胶原mRNA表达上调明显高于595nm激光,而1 064nm激光照射后兩种前胶原mRNA表达水平最高。

    Effects of Three Wavelength Laser Irradiation on the Expression of Procollagen Type Ⅰ and Ⅲ Procollagen mRNA in Cultured Fibroblasts

    Abstract: Objective In vitro cultured mouse fibroblasts were irradiated with 3 different wavelengths (595/755/1 064nm), and the effects of laser irradiation at different wavelengths on the expression of two types of procollagen (type I, type Ⅲ) in fibroblasts were observed and analyzed. Methods Neonatal mouse fibroblasts were cultured in vitro. According to the wavelength of laser irradiation, it was divided into 6 cases in control group, 6 cases in 595nm group with laser irradiation at 595nm, 6 cases in 755nm group with laser irradiation at 755nm and 6 cases in 1 064nm group laser irradiation at 1064nm. The expression of type I and type Ⅲ procollagen mRNA in fibroblasts was determined by RT-PCR. Results There were significant differences in the expression of procollagen mRNA among the four groups (P<0.05), and the relative expression of procollagen mRNA in 1 064nm group was the highest[ (113.07±2.85)×10-2,(118.72±1.96)×10-2,P<0.01],respectively. It was significantly higher than those in 595nm group [(96.74±1.03)×10-2,(101.51±1.64)×10-2,P<0.01],than those in 755nm group [(100.02±1.86)×10-2, (109.27±1.02)×10-2,P<0.01]. In the control group [(87.14±0.82) ×10-2,(100.48±0.73)×10-2,P<0.01]. The relative expression of typeⅠprocollagen mRNA in group B was significantly higher than that in control group (P<0.01), and the expression level of type Ⅲ procollagen mRNA in 755nm group was significantly higher than that in 595nm group and control group (P<0.01), but only typeⅠprocollagen mRNA in 595nm group was significantly higher than that in control group (P<0.01). Conclusion Laser irradiation can induce two precollagen (typeⅠ, type Ⅲ) mRNA expression in fibroblasts. The laser irradiation with a wavelength of 595nm can only promote the up regulation of mRNA expression of type I procollagen, and the up regulation of mRNA expression of type Ⅲ procollagen mRNA after 755nm laser irradiation is significantly higher than that of 595nm laser, while the highest expression level of two procollagen mRNA was observed after 1 064nm laser irradiation., 百拇医药(肖杰华 冶娟)
