细辛脂素联合受体脾细胞诱导心脏移植免疫耐受的研究(1)

     【摘要】目的:本实验通过术前7日和术前3日输注不同剂量供者脾细胞于受体胸腺内,联合中药细辛中的细辛脂素成分,探讨其对心脏移植免疫耐受的诱导,并对诱导效果加以比较。方法:提取中药细辛中的细辛脂素成分。健康雄性Wistar大鼠(供体)和SD大鼠(受体)各80只随机配对,建立大鼠腹部异位心脏移植模型,将接受心脏移植的SD大鼠分成5组:A组:阳性对照组,行腹部异位心脏移植,不给任何处置;B组:细辛脂素组:行腹部异位心脏移植,术前7天开始灌服细辛脂素25 mg·kg-1·d-1;C组:脾细胞诱导组1,术前7天开始输注供者脾细胞于受体胸腺内;D组:脾细胞诱导组2,术前7天开始输注供者脾细胞于受体胸腺内,术前7天开始灌服细辛脂素25 mg·kg-1·d-1;E组:脾细胞诱导组3,术前3天开始输注供者脾细胞于受体胸腺内,术前7天开始灌服细辛脂素25 mg·kg-1·d-1。每组各8只术后第7天处死。另外8只用于观察移植物生存天数。噻唑蓝(MTT)法检测供受体混合淋巴细胞反应;应用ELISA法检测受体血清中IL-2、IFN-γ、IL-4和IL-10水平; 结果:1、免疫诱导组联合细辛脂素组(D组和F组)移植心存活时间较阳性对照组显著延长(P<0.01)。2、供受体混合淋巴细胞反应免疫诱导组较阳性对照组差异有显著性(P<0.01)。3、免疫诱导组联合细辛脂素组(D组和F组)移植物生存时间和外周血细胞因子均有显著性差异(P<0.05)。结 论:1、通过术前7日胸腺内输注脾细胞可以诱导心脏移植免疫耐受,为供者特异性免疫耐受。2、术前7日预处理对心脏移植免疫耐受的诱导较术前3日预处理组理想。

    The Induction of Immune Tolerance in Rats Cardiac Allograft by Asarinin Combined with splenocyte

    【Abstract】Objective: To study the effect of immune tolerance in rats cardiac allograft induced by donor splenocyte 7 days and 1day before operation combined with Asarinin. Methods: To extract the asarinin from herba asari. The 80 SD rats (receptor) and 64 wistar rats (donor) were divided into five groups: group A (control group); group B (Asarinin treated 7 days before operation); group C (splenocyte treated 7 days before operation); group D (splenocyte treated 7 days before operation and Asarinin); group E (splenocyte treated 3 days before operation and Asarinin).Some were to examine survival time(n 8) and others were to observe mixed lymphocyte reaction(MLR) between the donor and receptor with MTT; the level of IL-2,IFN-γ,IL-4 and IL-10 in serum with ELISA postoperatively 7 days (n 8). Result: 1.Allografts survival time is prolonged in group D and E (P<0.01). 2.The lymphocyte proliferation is alleviated in group D and E comapred with group A(P<0.01). 3.The level of IL-4 and IL-10 significantly increases in group D and E compared with group A (P<0.01).
