    摘 要:目的:探讨溶栓颗粒对TNF-α损伤血管内皮细胞ICAM-1表达的影响。方法:用TNF-α(40ng/mL)损伤人脐静脉内皮细胞(HUVEC)造模,用含大、中、小剂量溶栓颗粒的大鼠血清干预受损HUVEC,应用HE染色观察细胞形态变化,免疫组化法观察血管细胞间黏附分子-1(ICAM-1)的表达。结果:TNF-α可以造成HUVEC损伤,表现细胞数量下降,皱缩、变形,分裂相细胞明显增加。受损细胞的ICAM-1表达明显增多。含大、中剂量溶栓颗粒的大鼠血清能减少ICAM-1表达,减少细胞有丝分裂。结论:溶栓颗粒可以减少因炎性介质TNF-α增加而受损的血管内皮细胞ICAM-1表达,或许可以减缓动脉粥样硬化形成;溶栓颗粒减少血管内皮细胞分裂增殖,对稳定动脉粥样斑块稳定性或许有积极作用。

    Effect of the Serum that Contain Rongshuan Grain on Injury

    Caused by TNF-α of the HUVEC on ICAM-1

    Abstract:Objective:To study the effect of Rongshuan Grain on Injury Caused by Tumor Necrosis Factor α(TNF-α) of the human umbilical vein endothelial cells(HUVEC) on the expression of intercellular adhesion molecule-1(ICAM-1). Methods: Use theTNF-α(40ng/mL)to damage the HUVEC, use the serum of Rats that contain different doses' Rongshuan Grain to interference the damaged HUVEC,and observe the morphological changes by HE staining and the expression of ICAM-1 by immunohistochemical method. Results: TNF-α can damage theHUVEC, manifestations included that the cell number is decreased,the cell shape change to crimple and thedivision cells areincreased.The damaged HUVEC'sexpression of ICAM-1 is increased.The serum of Rats that contain high dose and moderate dose of Rongshuan Grain can reduce the expression of ICAM-1, and reduce the number of division cells. Conclusion: Rongshuan Grain can reduce the expression of ICAM-1 on the damaged HUVEC by TNF-α. This function maybe decrease the formation of atherosclerosis. Rongshuan Grain can reduce the number of division cells of he damaged HUVEC,this function maybe effective on the stabilization of atheromatous plaque ......
