益肾壮骨颗粒剂对人工关节生物因素松动的影响(1)

    摘 要:目的: 探讨益肾壮骨颗粒剂对人工关节生物因素松动的影响。方法:随机选择准备行人工髋关节置换术患者20例。采集外周血,分离单个核细胞,每例标本分成5份。A组:仅单个核细胞,为正常对照组;B组:单个核细胞+骨水泥微粒,为模型组;C组:单个核细胞+骨水泥微粒+益肾壮骨低剂量组(3mg·mL-1);D组:单个核细胞+骨水泥微粒+益肾壮骨中剂量组(6mg·mL-1);E组:单个核细胞+骨水泥微粒+益肾壮骨高剂量组(12mg·mL-1)。培养48 h后,用ELISA测定细胞上清液中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量。结果:模型组的TNF-α和IL-6含量均明显高于正常对照组(P<0.01);单个核细胞+骨水泥微粒+益肾壮骨各剂量组的TNF-α和IL-6含量明显低于模型组(P<0.05)。结论:骨水泥微粒可刺激单个核细胞分泌TNF-α和IL-6,而益肾壮骨颗粒剂能有效地抑制单个核细胞分泌TNF-α和IL-6。益肾壮骨颗粒剂对人工关节生物因素松动方面有一定的防治作用。

    TheCytokine Study of Yishenzhuanggu'sEffect on the Artificial Joint Loosening

    Abstract:Objective:To explore the effects ofYiShenZhuangGu on aseptic cytokine of loosening of prothesis in vitro. Methods:From 20 patients,blood sample were taken, whose mononuclear cells were isolate and then divided into five groups.Group A:only mononuclear cells, which was control group;group B:mononuclear cells + Poly methyl methacrylate particles;Group C:mononuclear cells + Poly methyl methacrylate particles + YiShenZhuangGu (3mg·mL-1);Group D:mononuclear cells + Poly methyl methacrylate particles + YiShenZhuangGu (6mg·mL-1);Group E:mononuclear cells + Poly methyl methacrylateparticles + YiShenZhuangGu (12mg·mL-1).They were cultured for 48 hours, and the a mount of osteolytic factors such as TNF-a and IL-6 of each group was determined. Results: The levels of TNF-α and IL-6 were much higher in group B than A, C, D and E(P<0.01).The levels of TNF-α and IL-6 were much lower in group C, D and E than B(P<0.05).Conclusion: It shows the Poly methyl methacrylate particles could stimulate mononuclear cells to excrete TNF-α and IL-6, which can be inhibited by YiShenZhuangGu.By inhibiting bone resorption associated with the particles and TNF-α and IL-6, YiShenZhuangGu may have clinical utility in the prevention and treatment of aseptic loosening of prothesis. ......
