羊栖菜多糖对软脂酸诱导的HL-7702细胞凋亡的保护作用(1)

    摘 要:目的: 探讨羊栖菜多糖(SFPS)对软脂酸诱导的肝细胞凋亡的保护作用及其可能机制。方法:SFPS不同浓度(25、50、100μg/mL)预处理24h后,0.5mmol/L软脂酸(PA)作用48h诱导正常人肝细胞株(HL-7702)凋亡。MTT比色法检测SFPS对细胞增殖的影响;Annexin V-FITC标记法检测细胞的凋亡;RT-PCR法检测凋亡调控基因Bcl-2、Bax的表达并计算其比值(Bcl-2/Bax)。结果:0.5mmol/L PA处理48h的HL-7702细胞存活率下降为正常对照组的64.9%,细胞凋亡率为35.1%,Bax表达上升,Bcl-2表达及Bcl-2/Bax比值明显下降(均P<0.05);与模型组相比,SFPS不同浓度(25、 50、100 ug/ml)预处理后细胞存活率逐渐升高,细胞凋亡率显著降低,Bax表达逐渐下降,Bcl-2表达及Bcl-2/Bax比值逐渐上升(均P<0.05)。结论:SFPS对PA诱导的肝细胞凋亡有较好的保护作用,其作用机制可能与调节Bcl-2、Bax基因的表达有关。

    Protection of Sargassum Fusiforme Polysaccharides Against Palmitic

    Acid-induced Apoptosis on HL-7702 Cells

    Abstract:Objective: To investigate the protective effect of Sargassum Fusiforme Polysaccharides(SFPS) on HL-7702 cells against apoptosis induced by palmitic acid(PA) in culture, and then to explore the mechanism of SFPS against apoptosis.Methods: HL-7702 cells were exposed to PA to mimic apoptosis. To explore the effects of SFPS on the HL-7702 cells subjected to PA injury, cells were pretreated with SFPS 25, 50, 100μg/mL for 24h,respectively, and then exposed to 0.5mmol/L PA for 48h. The cell viability was measured by MTT colorimetric assay; Apoptotic cells were detected by Annexin V-FITC/PI double labeled flow cytometry;The expression of Bcl-2 mRNA , Bax mRNA and Bcl-2/Bax were checked up by Reverse transcription PCR(RT-PCR). Results: The results showed that PA treatment significantly decreased the cell viability, increased the percentage of apoptotic cells. Pretreatment of SFPS at different concentrations (25、50、100μg/mL)could remarkably increase the cell viability of hepatocytes. Compared with that of PA group (64.9±3.5)%, the cell viabilities of hepatocytes treated with 25,50 and 100 ug/ml SFPS increased to (70.0±3.0)%、(75.5±3.3)%、(84.1±3.7)% (P<0.05), respectively. The apoptotic rate of PA group was(39.8±4.7)%, while that of SFPS groups decreased significantly to(35.5±5.2)%、(28.2±3.3)%、 (17.1±2.8)% (P<0.05), respectively. Moreover, as detected by RT-PCR, the expression of Bax mRNA decreased, while that of Bcl-2 mRNA and Bcl-2/Bax increased significantly.Conclusion: These results suggest that SFPS has a protective effect on HL-7702 cells against apoptosis induced by PA , which might be related to regulating the expression of Bcl-2 and Bax in hepatocytes. ......
