黄芪注射液对重症肺炎大鼠胸腺细胞凋亡相关蛋白表达的影响(1)

     摘 要: 目的:探讨黄芪注射液对重症肺炎大鼠胸腺细胞凋亡相关蛋白表达的影响。方法:将40只Sprague-Dawley (SD)大鼠随机分为对照组、模型组、黄芪高剂量组、黄芪中剂量组和黄芪低剂量组,每组8只。采用气管穿刺注入感染法制作大鼠重症肺炎模型。分别采用原位末端转移酶标记法(Terminal deoxynucleotidyl transferase dUTP nick end labeling,TUNEL)检测各组大鼠胸腺细胞凋亡及Western blot检测凋亡相关蛋白cleaved caspase-3、Bcl-2和Bax的表达。结果:与对照组比较,模型组和黄芪治疗各组胸腺细胞凋亡增加,凋亡相关蛋白cleaved caspase-3和Bax表达增强,除黄芪高剂量组外Bcl-2表达均减弱, Bcl-2/Bax比值降低(P<0.05);与模型组比较,黄芪高剂量组胸腺细胞凋亡减轻,Bcl-2表达增强,cleaved caspase-3和Bax表达减弱(P<0.05),Bcl-2/Bax比值增加。结论: 黄芪注射液能减轻重症肺炎时胸腺细胞凋亡,其机制与促进Bcl-2表达,减轻Bax表达,增加Bcl-2/Bax比值有关。

    Influence of Astragalus Injection on Expressions of Apoptosis-associated Protein

    in Thymocytes of Rats with Experimental Severe Pneumonia

    Abstract:Objective:To investgate the influence of Astragalus injection on expressions of apoptosis-associated protein cleaved caspase-3,Bcl-2 and Bax in thymocytes of rats with experimental severe pneumonia. Methods:40 Sprague-Dawley (SD)rats were randomly divided into five groups: control group(n8), model group(n8),astragalus high dose group(n8) , astragalus medium dose group(n8) and astragalus low dose group (n8). The rat model of severe pneumonia was made by intratracheally instilling with Klebsiella pneumoniae suspension 0.3mL per animal. TUNEL method was used to assess the apoptosis of thymocytes. The expressions of cleaved caspase-3,Bcl-2 and Bax in thympcytes of rats with experimental severe pneumonia were detected with western blotting. Results:The severe pneumonia can result in the rise of apoptosis and the expressions of cleaved caspase-3 and Bax, and reduce the expressions of Bcl-2 of thymocytes and the ratio of Bcl-2/Bax. The changes can be improved significantly by high dose astragalus injection. Conclusion:Astragalus injection can significantly reduce the apoptosis and expressions of cleaved caspase-3 of thymocytes in rats with experimental severe pneumonia, and that may be closely associated with reducing the expressions of Bax and increasing the expressions of Bcl-2 and the ratio of Bcl-2/Bax of thymocytes in rats with severe pneumonia ......
