乌鳖颗粒对免疫性卵巢早衰小鼠CD+4/CD+8及(1)

     摘 要:目的:观察乌鳖颗粒对免疫性卵巢早衰小鼠CD+4/CD+8及Fas、FasL蛋白表达的影响。方法:采用雌性BALB/c小鼠皮下多点注射小鼠透明带多肽,建立免疫性卵巢早衰模型。将造模成功的小鼠随机分为模型组、己烯雌酚组、六味地黄丸组、乌鳖颗粒大、中、小剂量组。另设正常组和佐剂组,共灌胃给药4周。实验末应用免疫组化法测定脾脏CD+4、CD+8,计算CD+4/CD+8的比值。应用免疫组化法测定卵巢Fas、FasL蛋白的表达。结果:模型组小鼠脾脏CD+4/CD+8比值明显高于正常组和佐剂组(P<0.05)。乌鳖颗粒各剂量组小鼠脾脏CD+4/CD+8比值明显低于模型组(P<0.05,P<0.01)。模型组小鼠卵巢Fas蛋白表达低于正常组,Fas-L蛋白表达明显高于正常组,卵巢Fas、Fas-L蛋白表达和Fas/Fas-L比值与正常组比较具有显著性差异(P<0.05,P<0.01)。乌鳖颗粒大剂量组和中剂量组小鼠Fas蛋白表达明显高于模型组,(P<0.05,P<0.01)。乌鳖颗粒各剂量组Fas-L蛋白表达明显低于模型组(P<0.05,P<0.01)。乌鳖颗粒大剂量组、中剂量组小鼠Fas/Fas-L比值与模型组比较具有显著性差异(P<0.01)。结论:乌鳖颗粒对免疫性卵巢早衰小鼠具有调节CD+4/CD+8比值和Fas/FasL系统的作用。

    Effects of Wu Bie Granules on CD+4/CD+8and Fas/FasL Protein

    Expression of Mice with Immune Premature Ovarian Failure

    of Wu Bie granules on CD+4/CD+8and Fas/FasL protein expression of mice with immune premature ovarian failure.Methods: The zona pellucida 3 of mice was regarded as antigen,immune POF models of Balb /c female mice were established by multi-point injection of the antigen.The successful model of the mice were randomly divided into model group, diethylstilbestrol group, Liu Wei Di Huang Wan Group, Wu Bie granules of large, medium,low dose group,separate normal and adjuvant group.Each group began to be given the corresponding medicine for 4 weeks.End of the experiment,T lymphocyte cell subsets of the spleen were detected by immunohistochemistry;detected Fas, FasL protein expression of ovarian were also detected by immunohistochemistry.Results:The CD+4/CD+8 of Model group was significantly higher than that of the normal group and adjuvant group,showing significant difference (P<005).The CD+4/CD+8 of Wu Bie granules was significantly lower than that of the model group, compared with the model group,it was significant different(P<0.05,P<0.01). Fas protein expression of model group was lower than that of normal group;Fas-L protein expression was significantly higher than that of the normal group. Compared with the normal group Fas, Fas-L protein expression was significant difference (P<0.05,P<0.01).Wu Bie granules(large, medium dose) was significantly higher than that of Fas protein expression in model group. Compared with the model group it and significant difference (P<0.05,P<0.01).The expression of Fas-L protein of Wu Bie granules was significantly lower than that of the model group, compared with the model group it was significant different (P<0.05,P<0.01).Conclusion:Wu Bie granules can regulate CD+4/ CD+8 and the Fas / FasL system.

    卵巢功能早衰(premature ovarian failure,POF)是指女性40岁以前发生卵巢功能衰竭,表现为低雌激素,高促性腺激素的特征,并伴有面部潮红、潮热出汗、阴道干涩、头晕、情绪波动、失眠多梦及性欲减退等卵巢功能低落的临床表现。自身免疫异常是卵巢功能早衰的一个重要因素\[1\]。本文采用雌性BALB/c小鼠皮下多点注射小鼠透明带多肽,建立免疫性卵巢早衰模型。以探讨乌鳖颗粒对免疫性卵巢早衰小鼠CD+4/CD+8及Fas、FasL蛋白表达的影响。, http://www.100md.com(朱萱萱 王海丹 王进 施艳秋 刘志辉)
